摘要
目的探讨肺癌患者外周血单个核细胞(peripheral b lood mononuc lear cell,PBMC)中辅助性T细胞两个亚群(Th1/Th2)的功能状态及黄芪静脉应用对其的影响。方法采用逆转录聚合酶链反应(RT-PCR)方法测定了19例肺癌患者和10例健康对照者PBMC中Th1类细胞因子白细胞介素2(IL-2)和干扰素γ(IFN-γ)以及Th2类细胞因子IL-4、IL-6和IL-10的表达,并对肺癌患者与健康对照者及肺癌患者用黄芪前后上述指标进行比较。结果①19例肺癌患者IL-4、IL-6和IL-10的阳性表达例数分别为9、17和10,明显高于健康对照组,差别有统计学意义(P<0.05,P<0.01,P<0.05),IFN-γ和IL-2的阳性表达例数分别为1和2,与健康对照组比较,差别无统计学意义(P>0.05);②肺癌患者用黄芪后IL-4、IL-6和IL-10的阳性表达例数分别降至2、7和3,与用药前对比,差别有统计学意义(P<0.05),IFN-γ和IL-2的阳性表达例数升至7和11,与用药前比较,差别有统计学意义(P<0.05);③肺癌患者用黄芪后IL-2、IL-4、IL-6和IL-10的阳性表达例数与健康对照组比较,差别无统计学意义(P>0.05),IFN-γ的阳性表达例数与健康对照组比较,差别有统计学意义(P<0.05)。结论肺癌患者存在明显Th2优势应答,不利于机体抗肿瘤免疫;黄芪具有抑制肺癌患者PBMC Th2类细胞因子表达,增强Th1类细胞因子表达的作用,这可能是其抗肿瘤作用的机制之一。
Objective To explore the eytokine expressions of helper T lymphocyte(Th) subsets in peripheral blood mononuelear eells(PBMC) from lung cancer patients and the effects of Astragalus administered intravenously on it. Methods The expressions of interferon-γ(IFN-γ),interleukin-2(IL-2),IL-4,IL-6 and IL-10 in PBMC from 19 lung cancer patients and 10 healthy volunteers(control group) were tested by reverse transcription polymerase chain reaction (RT-PCR). IFN-γ and IL-2 are considered as Th1 eytokines. IL-4, IL-6 and IL-10 are considered as Th2 cytokines. Results (1)IL-4,IL-6 and IL-10 expressions in PBMC from 19 lung cancer patients(9,17 and 10) were significantly higher than those of control group( P 〈0.05, P 〈0.01, P 〈0.05). IFN-γ and IL-2 expressions in PBMC from 19 lung cancer patients(1 and 2) had no significant difference compared with those of control group( P ~0.05). (2)IL-4, IL-6 and IL-10 expressions in PBMC from 19 lung cancer patients decreased to 2,7 and 3 after using Astragalus,respeetively (all P 〈0.05). IFN-γ and IL2 expressions increased to 7 and 11 ,respectively( P 〈0.05). (3)IL-2,IL- 4,IL-6 and IL-10 expressions in PBMC from 19 lung cancer patients after using Astragalus had no significant difference compared with control group( P 〉 0.05). IFN-γ expressions had significant difference compared with that of control group ( P 〈0. 05). Conclusion There is a predominant expression of Th2 type cytokines in PBMC from lung cancer patients. Astragalus can down-regulate the Th2 type eytokine expressions and promote Thl-type eytokine expressions. This may be one of the antitumor effects of Astragalus.
