摘要
目的为了探讨虎杖与黄芪组方抗HSV时,其有效成分白藜芦醇与总皂苷之间的相互作用对药效的影响。方法以阿昔洛韦(ACV)为阳性对照,采用空斑减数实验,用中和作用原理对虎杖白藜芦醇与黄芪总皂甙合用抗HSV-2333株的药效进行了分析。结果在Hep-2细胞系统中,ACV对HSV-2333株增殖抑制的ED50为10.85ug/ml。当白藜芦醇与总皂甙以1(Dm白藜芦醇/1(Dm)总皂甙的比例合用时,对HSV-2333株直接杀灭、感染阻断、增殖抑制的ED50依次为0.41ug/ml,0.66ug/ml,0.86ug/ml;合用指数(CombinationIndex,CI)均<1.0,P<0.0005,说明这一合用比药效最强,协同作用最明显。结论ACV对HSV-2333株增殖抑制的治疗指数(Treatment Index,TI)为100。1(Dm)白藜芦醇/1(Dm)总皂甙对HSV-2333株直接杀灭、感染阻断、增殖抑制的TIs为248.3,154.2,118.4,为最佳合用比。
Objective:While resisting HSV with the formula of Rhizoma polygoni cuspidate and astragalus,whose effective components are the total saponin and resveratrol, the effect of the mutual action of which is explored on the pharmacodynamics,thereby we conducted the study. Methods:We take ACV as positive control,apply plaque reduction test,and make analysis on the pharmacodynamics in resisting HSV-2 333 strain by combining resveratrol of Rhizoma polygoni and total astragalin under TCM functional mechnism. Result: In the Hep-2 cell system, the ED50 of ACV in inhibiting the hyperplasia of HSV-2 333 strain was 10.85ug/ml. While combining in the ratio of resveratrol and total astragalin 1:1, the ED50 of direct removal,infection block and hyperplasia inhibition on HSV-2 333 strain was respectively 0. 41ug/ml,0. 66ug/ml and 0.86ug/ ml:their combination indexes(CI) were all less than 1.0, P〈0. 0005, this showed the combination of the ratio was strongest in pharmacodynamics with most marked cooperation. Conclusion:The treatment index(TI) was 100 in ACV resisting hyperplasia of HSV-2 333 strain. It was the best ratio of 1Dm resveratrol/1Dm total astragalin, where the Tis were respectively 248.3,154.2 and 118. 4 of direct removal,infection block and hyperplasis inhibition.
