摘要
目的:研究Brg1基因在胃腺癌组织中突变及蛋白表达意义,探讨其与肿瘤发展的关系。方法:应用聚合酶链反应及单链构象多态性(PCR-SSCP)技术检测37例胃腺癌和正常胃组织中Brg1基因第4和16外显子的基因突变,同时应用免疫组化技术分析30例胃癌组织B rg1蛋白的表达,初步探讨其和胃癌发生、发展及预后的关系。结果:37例胃腺癌标本中,Brg1第4和16外显子的基因突变率为零。B rg1蛋白在胃癌中的表达率为76.67%(23/30),与正常组织的表达率(44.44%)相比有明显增高,B rg1的表达与胃癌的浸润、淋巴结转移及分级分期均有关。结论:Brg1基因突变在胃癌中少见且不起主要作用,但其蛋白在胃癌中高表达并对胃癌的发生、发展起重要作用。B rg1的表达可作为判断胃癌恶性程度及预后的重要指标。
Objective : To investigate the mutation of Brgl gene and expression of Brgl protein in gastric adenocarcinoma , and to study its relation with tumor development. Methods:By means of polymerase chain reaction-single strand conform action polymorphism(PCR-SSCP) 37 cases of gastric mueosa were searched for Brgl gene mutation in exon 4 and 16. By immunohistochemistry the expression of Brgl protein in 30 cases of gastric adenocarcinoma was analyzed for their role in gastric cancer development and prognosis. Results : In 37 cases of gastric adenocarcinoma, no mutation of Brgl gene was detected. Brgl protein expression was 76.67% in gastric carcinoma which was higher than that in normal tissue 44.44%. The expression of Brg1 was related to tumor growth, lymphnode metastasis and tumor differentiation. Conclusion : Genetic abnormalities of Brgl might be rare and might not play a major role in stomach carcinogeneses. An increased expression of Brgl protein was associated with the advanced stage of gastric carcinoma, the Brgl expression can be used as an important indicator for differentiating malignancy and prognosis of gastric carcinoma.
