G protein is a superfamily of regulatory GTP hydrolases. They play pivotal roles in the signal transduction of cell growth and differentiation. Fluoride had been found to inhibit a variety of enzymes for more than half a century. Fluoride is a well-known G protein activator. The actual mechanism of fluoride action on guanine nucleotide-binding component of AC remained elusive. Many studies seem to suggest a correlation between fluorosis and G protein. The aim of this review is to discuss the basic chemical, biochemical, and toxicological properties of metallic fluoride, to understand the role of Al-F and Be-F complexes in human physiology and its potential effects on our health.
