生物素高产菌株摇瓶发酵条件优化研究 被引量:2

摘要 介绍了在摇瓶水平上对高产生物素菌株SporobolomgcesrosellsHK301进行摇瓶发酵条件优化,并获得S.cesrosellsHK301合成生物素的最佳发酵条件:葡萄糖为碳源,尿素为氮源,K2HPO41g/L;MgSO40.1g/L;FeSO40.01g/L。培养基初始pH值为7.2,接种量5%,最佳转速为230r/min。在此发酵条件下,生物素最高可达到540mg/L,生物素类物质产量为630mg/L,最终细胞干重为18.4g/L。 The effects of nutritional and environmental conditions on microbial biotin production by Sporobolorng cesrosells HK301 were investigated, Glucose was the optimum carbon source for the growth and biotin synthesized of S, cesrosells HK301, The use of inorganic nitrogen source stimulated biotin synthesis more efficiently than organic nitrogen sources, Compared with peptone and yeast extract, urea has been proved to be more efficient biotin production. The optimal concentration of urea was 20 g/L. The ratio of carbon to nitrogen (C/N) in the medium played central role on biotin production. Furthermore, it has been proved that high concentrations sulphur and ferrous iron were required for biotin production. The optimal nutritional and environmental conditions for biotin production by S. cesrosells HK301 were as following: glucose 10 g/L, urea 20 g/L, K2HPO4 1 g/L, MgSO4 0.1 g/L, FeSO4 0.01 g/L, inoculation temperature 30℃, initial pH 7.2 and 230 r/min. Subaequently, a batch culture was developed at the optimum culture time, and a maximum production of 540 mg/L biotin was achieved.
