雷公藤甲素对胶原诱导的关节炎大鼠关节局部热休克蛋白和主要组织相容性复合体Ⅰ类分子表达的影响 被引量:7

Effects of triptolide on expression of heat shock proteins and major hiscompatibility-Ⅰ molecule in synoviocytes and chondrocytes of collagen-induced arthritis rats

摘要 目的观察雷公藤甲素对胶原诱导的关节炎大鼠关节局部热休克蛋白(HSPs)和主要组织相容性复合体类分子(MHC-)表达的影响。方法建立胶原诱导的关节炎大鼠模型,雷公藤甲素按40μg/kg im给药,运用逆转录聚合酶链反应(RT-PCR)和免疫酶组织化学染色的方法,观察大鼠关节滑膜细胞和软骨细胞HSP60、HSP70和MHC-类分子的表达情况。结果与正常对照组相比,模型组大鼠关节滑膜细胞及软骨细胞HSP60、HSP70和MHC-类分子的表达均显著增高(P<0.05),与模型组相比,雷公藤甲素可以下调关节炎大鼠关节局部HSP60、HSP70和MHC-类分子的表达(P<0.05)。结论降低关节炎大鼠关节局部软骨细胞和滑膜细胞异常表达的HSPs与MHC-类分子的表达,可能是雷公藤甲素治疗类风湿关节炎(RA)的作用机制之一。 Objective To investigate the effects of triptolide on the expression of heat shock proteins(HSPs) and major histocompatibility-(MHC-) molecule in synoviocytes and chondrocytes of collagen-induced arthritis rats.Methods The experimental Wistar rat model of collagen-induced arthritis was es-tablished.The rats in triptolide group were administered with triptolide at 40μg/kg by im.The expres-sion of HSP60,HSP70,and MHC-molecule in synoviocytes and chondrocytes was detected by RT-PCRand immunohistochemical staining.Results To compare with normal group,the expression of HSPs andMHC-molecule increased remarkably in synoviocytes and chondrocytes of model rats(P<0.05).Trip-tolide could reduce the expression of these molecules in inflammatory joints rats(P<0.05).Conclusion Triptolide may play its curative role via down-regulating the exprssion of HSPs and MHC-molecule insynoviocytes and chondrocytes of rheumatoid arthritis(RA).
