摘要
贮脂细胞是目前肝纤维化研究的热点。本文参考Friedman等的不连续密度梯度离心法并作改良,建立了一种经济、简便、可靠的分离大鼠肝贮脂细胞、Kuffer细胞的方法。贮脂细胞得率约107/大鼠,纯度为90%以上,Kupffer细胞纯度约60%,其培养上清基本适合进一步研究用。本方法的建立,为进一步研究肝贮脂细胞与肝纤维化的关系,尤其是细胞水平及分子生物学水平的研究奠定了基础。
at-storing cells(FSC or Ito cells) have become
the focus in the study of liver fibrogensisat present. A modified simple and efficient density
gradient centrifugation method wasestablished in this paper. The yield rate was about l07 per
rat. The purity of FSC and Kupffercells (KC) was around 90%and 60, respectively. The survival
rate of the cells was about90%. This modified method of preparing FSC and KC may provide the
basis for the study ofliver fibrosis at the cellular and molecular level.
