摘要
目的:探讨重组人血小板源性生长因子(rh-PDGF-BB)和重组人碱性成纤维细胞生长因子(rhb-FGF)单独及联合应用对兔角膜基质成纤维细胞(RCF)增殖的影响。方法:采用细胞培养技术及噻唑蓝比色法(MTT)。结果:rhPDGF-BB和rhbFGF对RCF的促增殖作用较对照组有明显提高(P<0.01),rhPDGF-BB在10~100μg/L之间时对RCF有较强的促增殖作用(P<0.01),并呈剂量效应关系。rhb-FGF在0.1~10μg/L浓度范围内与RCF增殖呈剂量效应关系(P<0.01),当浓度增大到100μg/L,促增殖作用减弱。rhPDGF-BB和rhb-FGF联合应用时其促增殖作用较两种因子在相同浓度单独应用时有显著性差异(P<0.05)。结论:rhPDGF-BB,rhb-FGF单独或联合应用时有促进RCF体外增殖的作用。
· AIM: To evaluate the effects of recombinant human platelet derived growth factor-BB (rhPDGF-BB) and/or basic fibroblast growth factor (rhb-FGF) on the proliferation of cultured rabbit corneal fibroblasts.· METHODS: Rabbit corneal fibroblasts were exposed to rhPDGF-BB and/or rhb-FGF at various concentration for 1, 2, 3d respectively. The proliferation of the cells was studied with MTT assay.· RESULTS: The proliferating effects of cultured rabbit corneal fibroblast with either rhPDGF-BB or rhb-FGF were significantly enhanced than that of the control group at 24, 48 and 72h (P <0.01). RhPDGF-BB at 10-100μg/L or rhbFGF at 0.1-10μg/L promoted RCF growth in a dose and time dependant way (P <0.01). The proliferating effects of rhb-FGF at 100μg/L on cultured rabbit corneal fibroblast decreased. RhPDGF-BB at 10μg/L and rhb-FGF at 0.1μg/L showed stronger effects on cultured rabbit corneal fibroblasts growth than both used individually (P <0.05).· CONCLUSION: RhPDGF-BB and/or rhb-FGF can promote the proliferation of the cultured rabbit corneal fibroblast.·
