摘要
目的 通过材料浸渍液法测定兔角膜成纤维细胞的相对增殖度来评价微孔和无孔聚甲基丙烯酸 β 羟乙酯(PHEMA)的生物相容性。方法 用消化法原代培养兔角膜成纤维细胞 ,分别将微孔PHEMA和无孔PHEMA浸入细胞培养基 2 4h并用此浸渍液培养兔角膜成纤维细胞 ,分别于 2、4、7d后用分光光度计测定吸光度并计算细胞相对增殖度。结果 微孔PHEMA组及无孔PHEMA组的细胞相对增殖度均大于 80 % ,细胞毒性为一级。
ObjectiveTo evaluate the biocompatibility of porous PHEMA and non-porous PHEMA by measuring corneal fibrolasts cell relativ e growth rate(RGR) in rabbit.MethodsPrimary corneal fibroblasts of rabbit wer e cultured by digestion method.Porous PHEMA and non-porous PHEMA were separatel y impregnated in nutrient medium for 24 h and corneal fibroblasts of rabbit wer e subcultured with the media.At 2,4 and 7 days,light absorbance was mensurated by spectrophotometer to evaluate the relative growth rate of corneal fibroblast s.ResultsRGRs of corneal fibroblasts in both groups were over 80%,and the cy totoxicities were Ⅰgrade.ConclusionPorous PHEMA and non-porous PHEMA have less influence on the growth of rabbit corneal fibroblasts.
