摘要
目的检测知母宁体外抗单纯疱疹病毒Ⅱ型(HSVⅡ)作用。方法以Vero细胞为宿主细胞,阿昔洛韦为阳性对照药物,采用中性红染色法测定不同药物加入方式下知母宁的体外抗HSVⅡ有效率,并检测其抑制病毒作用的时效关系及对病毒感染性的影响。结果知母宁表现出对HSVⅡ333的综合抑制作用和抑制病毒吸附后的后续复制增殖作用,其抗病毒有效率(ER%)最高达80%,这两种作用的半数有效浓度(EC50)均约为0.8mg/ml。知母宁浓度≥2.08mg/ml时能显著抑制HSVⅡ,ER%最高达95%。知母宁还有较弱的预防病毒吸附作用。知母宁抗病毒作用的时效关系研究表明,随药物作用时间延长,低浓度药物抗病毒有效率呈减小趋势,在高浓度时(≥2.08mg/ml)其病毒抑制强度基本维持稳定。随药物作用时间延长,知母宁可明显削弱病毒的感染性,知母宁组病毒的-lgTCID50值较病毒对照组的下降值呈增大趋势,其下降百分比维持稳定,约为40%。结论知母宁体外抗单纯疱疹病毒Ⅱ型效果好,且其抗病毒作用具有多个作用点。
Objective To study the anti-viral effect of chinonin against HSV-Ⅱ in vitro.Methods The anti-viral effective rate (ER%) of chinonin with different drug addition methods against HSV-Ⅱ 333 was assayed in vitro by neutral red staining method, with Vero cell as host cell and acyclovir as positive reference drug. Also, the time-effect relationship of its anti-viral effect and its influence on virus infectivity was studied.Results Chinonin was demonstrated to have comprehensive inhibitive effects on HSV-Ⅱ 333 as well as inhibition on the proliferation phase after virus adsorption, with the highest anti-viral ER% about 80%. The median effective concentration (EC_(50)) of both effects was about 0.8 mg/ml. Chinonin was also confirmed to have remarkable direct inhibition against HSV-Ⅱ at concentration ≥2.08 mg/ml and its maximal ER% reached 95%. It displayed weak preventive effect against the virus adsorption. The time-effect relationship of anti-viral effect of chinonin indicated that the anti-viral ER% of chinonin was decreased with the prolongation of drug treating time at low concentration, however, the drug showed a basically stable inhibitive intensity to the virus at concentration ≥2.08 mg/ml. With the prolongation of drug treating time, chinonin was demonstrated to greatly weaken the infectivity of HSV-Ⅱ, and the drop of -lg TCID_(50) value of chinonin group (compared with that of virus control group) showed an increased tendency with a stable drop percentage about 40%. Conclusion Chinonin showed good anti-viral effect against HSV-Ⅱ in vitro. It inhibited the virus at several effect points.
