摘要
目的:研究中药五环三萜类有效成分熊果酸和齐墩果酸抗人肺癌细胞增殖、侵袭的作用,并探讨其抗侵袭作用的机理。方法:用熊果酸和齐墩果酸分别作用于克隆化高转移人肺癌细胞(PGCL3),测定细胞增殖抑制率、软琼脂集落形成率、对重建基底膜的侵袭能力、趋化运动能力、对层粘连蛋白的粘附能力以及组织蛋白酶B活性分泌等多项指标。结果:熊果酸和齐墩果酸均可降低PGCL3细胞增殖能力(P<0.001),IC50分别为44.73μmol/L和40.71μmol/L;30μmol/L、40μmol/L熊果酸和35μmol/L、45μmol/L齐墩果酸均能抑制PGCL3细胞的侵袭能力(P<0.001)且有剂量依赖性。抗侵袭作用机理的分析结果显示上述浓度的两种药物对细胞的运动、粘附能力及组织蛋白酶B分泌均有明显的抑制作用且均有统计学的显著意义,软琼脂集落形成率也显著降低。结论:熊果酸和齐墩果酸均有抗人肺癌细胞增殖和侵袭能力,其抗侵袭作用不是对侵袭的某一环节的阻断,而是对侵袭各个基本环节的全面抑制作用。
Objective :To investigate the effects of Ursolic acid (UA ) and oleanolic acid(OA) , both the active components of pent cyclic triterpene acid, on proliferation inhibition, anti - invasive activity in highly metastasis lung cancer cell line (PGCL3) ,and to make some analysis on their anti -invasion mechanism. Methods: We tested the cell proliferation inhibition rate, colony -formation rate in soft agar ,the invasive ability wit matrigel reconstituted basement membrane model, the chemo-tatic migration activity, the laminin adhesion ability and the secretion activity of cathepsin B after treatment of UA and OA in PGCL3 cell Iine respectively. Results : Both UA and OA can decrease the cell proliferation rate significantly, and the IC50 value is 44.73μmol/L and 40.71μmol/L respectively, and 30μmol/L and 40μmol/L UA, 35μmol/L and 45μmol/L OA can repress obviously the invasive ability of PGCL3 cell line in a dose - dependent manner. We made some analysis and found which made inhibitive effect to the migration ability, adhesion ability and the secretion of cathpsin B in a concentration - dependent manner, also the colony -formation rate in soft agar. Conclusions: Both two drugs can inhibit the proliferation ability and invasive ability, it showed that acted on cell not through any one point only of invasion but likely through all.
