宁痫冲剂对戊四唑致痫大鼠脑源性神经营养因子蛋白表达的影响 被引量:2

Effect of Ningxian Granule on Brain-derived Neurotrophic Factor Protein Expression in Rats with Pentylenetetrazol- Induced Epilepsy

摘要 [目的]探讨以升清降浊法组成的中药复方宁痫冲剂的抗痫作用机理。[方法]SD大鼠40只,随机分为5组:即正常组,模型组,宁痫冲剂高、低剂量组(10g/kg和5g/kg)及苯巴比妥组(60 mg/kg);除正常组外,其他4组均以戊四唑(PTZ)亚惊厥剂量35mg/kg诱导慢性癫痫模型,造模同时各组按设定剂量分别给药,1次/d,连续4周;动物处死后以免疫组织化学方法,观察宁痫冲剂对大脑额叶皮质、海马CA3区脑源性神经营养因子(BDNF)蛋白表达的影响。[结果]模型组大鼠脑内海马、皮质区免疫标记阳性神经元数目均增多(P<0.05或P<0.01),标记强度增强;宁痫冲剂低剂量组能减少免疫阳性神经元数目(P<0.05或P<0.01),减弱免疫反应性。[结论]PTZ慢性诱导癫痫反复发作可导致脑内BDNF表达增强;宁痫冲剂的抗痫作用机制可能与降低BDNF蛋白表达有关。 [ Objective ] To investigate the mechanism of Ningxian Granule (NG), a compound prescription with lucidity-ascending and turbidity-descending actions. [ Methods ] Forty SD rats were randomized into 5 groups: A (normal), B (model), C (low-dose NG, 5 g/kg), D (high-dose NG, 10 g/kg) and E (phenobarbital, 60 mg/ kg). Except Group A, the others were epileptic rats induced by pentylennetetrazol (PTZ, 35 mg/kg). NG and phenobarbiatal were given to different groups accordingly, once a day for 4 weeks. The effects of NG on brain-derived neurotrophic factor (BDNF) protein expression was observed by using immunohistochemistry technique. [Results] The number of positive neuron increased both in the hippocampus and cerebral cortex of epileptic rat brain (P < 0.05 or P < 0.01) and label intensity increased; the number of positive neuron decreased in low-dose NG group (P < 0.05 or P < 0.01), and NG reduced BDNF-immunoreactivity. [Conclusion] PTZ-induced epilepsy can increase BDNF expression; the mechanism of anti-epileptic effect of NG might be related with the reduction of BDNF protein expression.
