急性白血病全血样品的质子激发X荧、光分析方法学研究 被引量:3

Study on PIXE for Analysis of Trace Elements in Whole Blood of Patients with Acute Leukemia

摘要 作者对AXIL程序改进后编写的多功能X射线谱分析程序PAXMF能有效地分解复杂X射线能谱。具体给出了用相对灵敏度因子的内标法确定样品中各元素(绝对或相对)含量的计算公式。本文还介绍了供PIXE法用的全血样品的采集及薄靶制备的步骤和方法。采用厚度为100μm的奇特络吸收片可使X射线的能谱质量得到改善,使可分析的元素数目达到11个。最小探测限可达到0.05~1.0ppm或10^(-8)~10^(-10)g。分析结果表明,急性白血病患者全血中Cu、Ca、P、S、Si的含量及Cu/Zn比值都明显高于正常人,而病人全血中Zn、K、Fe、Al、Rb五种元素的含量则比正常人明显偏低。结果表明,Cu,Cu/Zn值是有用指标,Al和Rb可能是在急性白血病中起重要作用的元素。 A computer program PAXMF with multi-functions, which is a modified AXIL programm,has been provided to efficiently to resolve the complex X-ray spectra. A formula is deduced to calculate the contents (absolute or relative) of some elements in a sample by internal standard method with the aid of relative sensitivity factors. The collection of whole blood sample and the procedures of thin target preparation for PIXE are also described. A Cr funny foil with thickness of 100μm as an absorber is placed in front of the Si(Li) detector. It is very efficient in improving the quality of the measured X-rey spectra. The number of analysed elements is increased up to 11 (Al, Si, P,S, K, Ca, Mn, Fe, Cu, Zn and Rb) in the present work with minimum detection limit (MDL) of 0. 05~l. 0μg/g or 10~10μg. Some significant results have been obtained with the analysis of PIXE spectra and data statistical processing. The concentrations (in μg/g, dry weight) of Cu, Ca, P, S, Si and the radio of Cu to Zn, are significantly higher and those of Zn, K, Fe, Al and Rb are significantly lower in acute leukemia patients than those in normal controls. The results show the content of Cu and the ratio of Cu to Zn may be as valuable indices of the extent for disease and the elements of Al and Rb may play an important roles in acute leukemia etiology.
