摘要
目的研究大豆异黄酮对转基因MMTV-erbB-2小鼠的乳腺癌相关激素及受体效应。方法选择20~22 g的雌性转基因MMTVerbB-2小鼠80只,其中40只给予大豆异黄酮进行饲养作为观察组,另40只给予普通饲料作为对照组。比较两组小鼠体重、肿瘤发生率、潜伏期;检测比较两组小鼠未发生肿瘤乳腺组织中睾酮(T)、雌二醇(E2)、泌乳素(PRL)、促黄体生成素(LH)和促卵泡激素(FSH)的含量,雌激素受体(ER)、孕激素受体(PR)、增殖细胞核抗原(PCNA)阳性表达率。结果观察组小鼠体重下降与对照组比较差异有统计学意义(P【0.01);观察组与对照组肿瘤潜伏期无明显差异(P】0.05);观察组肿瘤发生率低于对照组(P【0.01);观察组的E2、T、PRL水平显著高于对照组(P【0.05);观察组的FSH和LH水平显著低于对照组(P【0.01);两组ER、PR受体阳性率差异无统计学意义(P】0.05);观察组PCNA阳性率大于对照组,差异具有统计学意义(P【0.05)。结论大豆异黄酮可以降低转基因小鼠MMTV-erbB-2的肿瘤发生率,提高PCNA阳性表达率。
Objective To analyze the soy isoflavones on breast cancer-related hormones and receptor effects in genetically modified MMTV-erbB-2 mice. Methods 80 female transgenic MMTV-erbB-2 mice of 20 ~ 22 g were selected. 40 of which were given soy isoflavones as the observation group,and the other 40 were given normal diet as control group. Body weight,tumor incidence and latency of two groups were compared. The testosterone( T),estradiol( E2),prolactin( PRL),luteinizing hormone( LH) and follicle-stimulating hormone( FSH) levels,estrogen receptor( ER),progesterone receptor( PR),proliferating cell nuclear antigen( PCNA) expression rate in two groups were detected and compared. Results The decreased body weight of observation group was statistically significant than that of control group( P< 0. 01). There was no significant difference in incubation period between two groups( P>0. 05). The incidence of tumors of the observation group was statistically significance than that of control group( P<0. 01). E2,T and PRL levels were significantly higher in observation group than those of control group( P < 0. 05); FSH and LH levels in observation group were significantly lower than those of control group( P < 0. 01). The positive rate of ER,PR showed no significant difference in two groups( P>0. 05). The PCNA-positive rate of observation group was statistically significant higher than that of control group( P<0. 05). Conclusions Soy isoflavones can reduce the incidence of tumors in transgenic MMTV-erbB-2 mice and increase the expression of PCNA-positive rate.
