摘要
比较了5种大孔树脂对金丝草(Pogonatherum crinitum)中总黄酮的吸附及解吸性能,采用静态和动态的吸附-解吸试验,利用紫外可见分光光度计测量金丝草总黄酮的浓度,研究不同的工艺条件对总黄酮纯化的影响。结果表明,静态分离纯化工艺的最佳参数为DA-201大孔树脂质量与吸附液体积的比值1∶10(m/V,g∶mL)、吸附液pH 3.00、恒温振荡时间3 h、解吸液为70%的乙醇溶液。动态分离纯化工艺的最佳参数为采用2 mL/min流速上样,用3个柱体积的70%乙醇,以2 mL/min流速洗脱可较好地分离纯化金丝草总黄酮。
Absorbability and desorption property of five macroporous resins for flavonoids from Pogonatherum crinitum was compared using static and dynamic absorption / desorption methods;and the purification process was evaluated by measuring the concentration of total flavonoids with UV spectrophotometer.The results showed that the best technical parameters of static adsorption were,ratio of DA-201 macroporous adsorption resins to extractant,1∶10(m / V,g∶mL);pH,3.00;time for shaking at constant temperature,3 h;and 70% alcohol as desorption solution.For dynamic purification,the optimum technical parameters were as follows,current velocity of adsorption,2 mL / min,volume of desorption solution(70% ethanol),3 BV;and elution velocity,2 mL / min.
