摘要
目的初步探讨信号转导子和转录激活子STAT3在宫颈癌发生发展中的作用以及槲皮素对宫颈癌细胞中STAT3表达的影响。方法用RT-PCR检测不同浓度的槲皮素作用人宫颈癌HeLa细胞中STAT3 mRNA的表达,并以细胞免疫荧光技术观察凋亡细胞内STAT3和磷酸化STAT3(P-STAT3)的改变。结果槲皮素处理HeLa细胞后,当槲皮素浓度从6.25μmol/L增加到50μmol/L,相同时间HeLa细胞内STAT3 mRNA表达逐渐降低,除6.25μmol/L组与溶剂对照组相比无统计学差异(P>0.05)外,余差异全部有统计学意义(P<0.05),且处理24h后,STAT3 mRNA表达量与给药浓度呈负相关,r1=-0.861,P=0.000;各时间段不同浓度组(6.25、12.5、25、50μmol/L)两两相比,差异均有统计学意义(P<0.05)。随着时间变化,相同浓度组中STAT3 mRNA表达明显降低,在给予6.25μmol/L槲皮素后,STAT3 mRNA的表达量呈时间依赖性,r2=-0.539,P=0.000。运用免疫荧光技术观察到STAT3蛋白及磷酸化水平随槲皮素浓度及时间增加表达呈明显下降趋势。结论槲皮素能明显抑制HeLa细胞的生长;槲皮素可抑制细胞内STAT3表达,从而可能通过抑制JAK/STAT信号通路诱导宫颈癌细胞凋亡。
Objective To discuss the STAT3's role for development of cervical carcinoma and the effect of quercetin for STAT3 expression on cancer cells.Methods The expressions of STAT3 mRNA with different concentrations of quercetin were detected by RT-PCR and STAT3 and phosphorylation of STAT3 (P-STAT3)in human cervical carcinoma HeLa cells were observed by immunofluorescence.Results When the quercetin concentration varied 6.25 μmol/L from 50 μmol/L,STAT3 mRNA was decreased gradually in HeLa cells,and expression of STAT3 mRNA in different concentrations was significantly lower compared with the DMSO control group excluding the group of 6.25 μmol/L in the same time,the differences were statistically significant (P<0.05), and for 24 hours,STAT3 mRNA expression was negatively correlated with the dose levels,r1=-0.861,P=0.000;different concentrations (6.25,12.5,25,50 μmol/L)compare with each other in the each time,the differences were statistically significant(P<0.05).As time changes,in the same concentration group,STAT3 mRNA expression was lower,in the group of 6.25 μmol/L,STAT3 mRNA expression was a time-dependent manner,r2=-0.539,P=0.000.STAT3 protein and its phosphorylation levels were significantly reduced by immune fluorescent technique.Conclusions Quercetin can inhibit the growth of HeLa cells;quercetin may inhibit the expression of STAT3 in cells,and induce the apoptosis of cervical cancer cells by inhibiting the JAK/STAT signaling pathway.
