摘要
目的探讨苦参碱(matrine,Ma)和奥沙利铂(oxaliplatin,L-OHP)联合应用对人结肠癌细胞SW1116的增殖抑制作用及机制。方法用不同浓度的Ma、L-OHP和两药联用处理人结肠癌细胞SW1116,采用MTT法测定细胞的增殖抑制作用,TUNEL法检测细胞凋亡率,半定量RT-PCR方法检测fas和fasL基因mRNA表达水平。结果Ma、L-OHP单独应用和联合应用均能抑制SW1116增殖,Ma和L-OHP联合应用后对人结肠癌SW1116细胞的抑制率明显大于单药,并有显著的协同杀伤作用。与单药相比,联合用药组的细胞凋亡率显著上升(P<0.01),Fas mRNA表达显著上升(P<0.01),FasL mRNA表达明显下降(P<0.01)。结论Ma和L-OHP联合应用具有明显的协同抗结肠癌SW1116细胞增殖的作用,并能诱导细胞凋亡,其机制可能与上调fas基因和下调FasL基因表达有关。
ObjectiveTo investigate the inhibitory effects and mechanism of Matrine combined with Oxaliplatin on human colon cancer cell line SW1116.
