高海拔地区地塞米松对大鼠脑损伤后脑水肿与水通道蛋白之间表达影响的研究

Study on the Effect of Dexamethasone on Expression of Aquaporin 4 after Brain Injury in Rats at High Altitude Area

摘要 目的:探讨高海拔地区SD大鼠脑损伤后脑水肿与水通道蛋白(AQP4)之间的关系,并观察常规剂量地塞米松对脑损伤后脑水肿的效果,为西宁地区颅脑外伤后脑水肿的发生、发展及常规剂量地塞米松治疗脑水肿提供理论依据和实验基础。方法:将144只SD大鼠随机分为4组:空白组、假手术组、对照组及地塞米松组,每组各36只,制备SD大鼠重型颅脑损伤模型,并使用地塞米松进行治疗,分别于不同时间点对模型大鼠进行灌注取脑,对各组模型大鼠脑组织进行免疫组化染色,观察各组大鼠脑水肿变化情况,同时收集实验相关数据。结果:对照组大鼠脑组织AQP4表达累积光密度值高于地塞米松组(P<0.05),明显高于假手术组及空白组(P<0.05)。地塞米松组大鼠脑组织AQP4表达累积光密度值高于假手术组及空白组(P<0.05)。假手术组与空白组比较差异无统计学意义(P>0.05)。对照组与地塞米松组的大鼠脑组织标本于伤后AQP4表达上调,且对照组于24h、72h及168h的表达水平较高。结论:高海拔地区SD大鼠重型颅脑损伤模型的脑水肿高峰期为伤后72h,伤后168h恢复至低于24h水平;高海拔地区SD大鼠重型颅脑损伤模型脑水肿的进展过程与AQP4表达的变化规律呈正相关;高海拔地区地塞米松对SD大鼠重型颅脑损伤模型的脑水肿有缓解作用,对AQP4的表达有负性调节作用。 Objective:This paper is aimed tostudy the relationship between brain edema and aquaporin(AQP4)after brain injury in SD rats in high altitude in order to observe the effect of routine dexamethasone on brain edema after brain injury.This study provides theoretical basis and experimental basis for the occurrence and development of brain edema after brain injury in Xining area,and for the treatment of brain edema with routine dose of dexamethasone.Methods:SD rats were randomly divided into 4 groups:Blank group,sham group,control group and dexamethasone group,each of which has 36 rats.Preparing SD rat model of severe head injury and using dexamethasone for treatment,the cerebral perfusion in rats was taken respectively at different time points;the model for each model immunohistochemical staining in tissue of rats was applied;their changes were observed between groups brain edema andrelevant experimental data were collected.Results:The cumulative optical density of AQP4 expression in brain tissue of rats in control group was higher than that in dexamethasone group(P<0.05),which was also higher than that in sham operation group and blank group(P<0.05).The cumulative optical density of AQP4 expression in dexamethasone group was higher than that in sham operation group and blank group(P<0.05).There was no significant difference between the sham operation group and the blank group(P<0.05).The expression of AQP4 was upregulated in brain tissue specimens of the control group and dexamethasone group after injury,which was higher in the control group at 24 h,72 h,and 168 h at each time point.Conclusion:The peak of brain edema in severe craniocerebral injury of the model of SD rats in high altitude area is 72 h after injury,and its recovery lowered to 24 h from 168 h after injury.There was a positive correlation between the progression of brain edema and the expression of AQP4 in SD rats with severe craniocerebral injury at high altitude.Dexamethasone at high altitude can relieve the brain edema of SD rats with severe craniocerebral injury and can regulate the expression of AQP4 negatively.
