摘要
目的:探讨2-甲氧基雌二醇(2-ME)对骨肉瘤移植瘤的抑制作用、可能的分子机制及不良反应。方法:构建裸鼠皮下骨肉瘤模型,2-ME作用后,记录肿瘤的体积和质量;通过移植瘤HE、免疫组化、TUNEL染色,观察肿瘤组织的增殖抑制、诱导凋亡及Bcl-2、VEGF、Caspase-3的表达情况,检测2-ME对裸鼠肝肾功能的影响。结果:2-ME对肿瘤的体积和重量均有抑制作用,HE染色显示,实验组出现不同程度的肿瘤细胞密度减低;免疫组化结果表明,随着2-ME浓度的升高,肿瘤组织内Bcl-2、VEGF的表达逐渐下降,而Caspase-3的表达则逐渐升高;TUNEL结果表明2-ME诱导裸鼠移植瘤组织细胞凋亡;肝肾功能检测未见损害发生。结论:2-ME在体内能有效抑制骨肉瘤MG63细胞的增殖、诱导其凋亡,其抗骨肉瘤作用机制与Bcl-2、VEGF、Caspase-3相关,且未见明显毒副作用。
Objective:To investigate the inhibitory effect,potential molecular mechanism and adverse reac-tions of 2-methoxyestradiol(2-ME)on osteosarcoma xenografts.Methods:Osteosarcoma subcutane-ous tumor model was established by using osteosarcoma MG63 cells.The volume and weight of tu-mor were recorded after 2-ME treatment.The proliferation inhibition and apoptosis of tumor tissue were observed by HE,immunohistochemistry and TUNEL staining.The expression of Bcl-2,VEGF and Caspase-3 was detected,and the effect of 2-ME on liver and kidney function in nude mice was examined.Results:2-ME inhibited the volume and weight of tumors.HE staining showed that the experimental group had different degrees of tumor cell density reduction.The results of immuno-histochemistry showed that the expression of Bcl-2 and VEGF was decreased gradually with increas-ing 2-ME doses,while the expression of Caspase-3 increased gradually.TUNEL results showed that2-ME induced apoptosis of tumor cells in nude mice.No liver and kidney function damage was detect-ed.Conclusion:2-ME can effectively inhibit the proliferation and induce apoptosis of osteosarcoma MG63 cells in vivo.The anti-osteosarcoma mechanism is related to Bcl-2,VEGF and Caspase-3.In addition,there are no obvious side effects after 2-ME treatment.
