摘要
目的观察电针干预"足三里""关元"对类风湿性关节炎模型大鼠的治疗效果及血清肿瘤坏死因子α(tumor necrosis factor alpha,TNF-α)和免疫球蛋白M(immune globulin M,IgM)的调控作用。方法SD大鼠24只,随机分成空白组、模型组、电针组。采用胶原诱导关节炎(collagen induced arthritis,CIA)造模方法制备类风湿性关节炎大鼠模型,持续20 d,造模成功后,对电针组大鼠进行电针"足三里""关元"刺激。1次/d,15 min/次,5 d/疗程,共5个疗程。观察大鼠一般状态,每隔3 d测量大鼠前、后肢腕踝关节直径并进行关节炎指数(arthritic index,AI)评分、测量体质量,ELISA法检测大鼠血清TNF-α、IgM含量水平,HE染色观察大鼠踝关节组织病理学改变。结果与空白组相比,模型组大鼠左右踝关节直径、AI评分、血清TNF-α、IgM表达水平均显著升高(均P<0.01);与模型组相比,电针组大鼠左右踝关节直径、AI评分、血清TNF-α表达水平均显著降低(P<0.01,P<0.01,P<0.05);HE染色显示模型组大鼠踝关节关节滑膜细胞增生,关节软骨破坏,关节腔消失且大量炎性细胞浸润,电针组大鼠踝关节损伤较模型组明显减轻。结论电针治疗能够减轻类风湿性关节炎大鼠踝关节的肿胀程度及病理损伤,其作用机制可能与抑制TNF-α炎症因子相关。
Objective To observe the curative intervention“foot three mile”and Guanyuan(RN4)effect on the treatment of rheumatoid arthritis model in rats and the serum tumor necrosis factorα(TNF-α)and immune globulin M(IgM)regulation.Methods SD rats 24,randomly divided into blank group,model group,electric acupuncture group.Collagen induced arthritis(CIA)building,method of preparation of rheumatoid arthritis rats model for 20 d,after the success of the building,electric acupuncture on the curative group rats foot Zhusanli(ST36)and Guanyuan(RN4).1 time/d,15 min/time,5 d/treatment,a total of five cycles.To observe the general condition of rats,every 3 d measuring wrist ankle diameter front and hind legs of rats and arthritis index(AI)score,measuring weight,ELISA method to detect the rat serum TNF-α,IgM levels,HE dyeing observation ankle histopathological changes in rats.Results Compared with the blank group,model group rats left ankle diameter,AI,serum TNF-α,IgM expression levels were significantly higher(P<0.01);Compared with model group,the curative group rats left ankle diameter,AI,serum TNF-αexpression levels were significantly lower(P<0.01;P<0.01;P<0.05);HE staining showed model group rats ankle joint synovial cell hyperplasia,articular cartilage damage disappeared,articular cavity and a large number of inflammatory cells infiltration,ankle injury in rats electric acupuncture group compared with model group significantly reduce.Conclusion The electric acupuncture treatment can reduce rheumatoid arthritis rats ankle swelling degree and the pathological damage,its mechanism may be related to the inhibition of TNF-αinflammation factors.
