摘要
【目的】探讨五味子提取物的抗抑郁作用及机制。【方法】取40只大鼠建立慢性不可预测轻度应激(CUMS)抑郁模型,随机分为模型组,五味子提取物低、中、高剂量组,每组各10只,另设正常组10只。相应干预结束后,进行旷场实验和蔗糖偏好实验评估大鼠行为学。处死大鼠后,尼氏染色法观察海马神经元形态变化,原位末端标记法(TUNEL)染色测定海马神经元凋亡率,Western Blot法检测海马组织Janus蛋白酪氨酸激酶1(JAK1)、信号传导及转录活化因子3(STAT3)及磷酸化JAK1(p-JAK1)、磷酸化STAT3(p-STAT3)蛋白相对表达量。【结果】与模型组比较,五味子提取物各剂量组水平运动得分、垂直运动得分、1%蔗糖水消耗率、p-JAK1和p-STAT3蛋白相对表达量升高,海马神经元凋亡率降低(P<0.05),其中五味子提取物高剂量组上述指标改变最明显(P<0.05)。尼氏染色结果显示,模型组海马神经元萎缩、排列紊乱稀疏,坏死细胞较多,而五味子提取物各剂量组均较模型组有不同程度改善。【结论】五味子提取物有显著的抗抑郁作用,其机制可能与激活JAK1/STAT3信号通路从而发挥保护海马神经元有关。
Objective To explore the anti-depressant effect and mechanism of Fructus Schisandrae Chinensis extracts.Methods Forty rats were selected to establish chronic unpredictable mild stress(CUMS)depression model,and then were randomly divided into model group,low-dose,medium-dose and high-dose of Fructus Schisandrae Chinensis extracts groups,with 10 rats in each group,and 10 rats in normal group.After corresponding intervention ending,open field test and sucrose preference test were performed to evaluate the ethology in rats.After the rats were sacrificed,the morphological changes of hippocampal neurons were observed by Nissl’s staining,the apoptosis rate of hippocampal neurons was analyzed by terminal deoxy(d)-UTP nick end labeling(TUNEL),and the relative protein expression levels of Janus kinase 1(JAK1),signal transduction and transcription activation factor 3(STAT3),phosphorylation JAK1(p-JAK1)and phosphorylation STAT3(pSTAT3)were detected by Western Blot assay.Results Compared with the model group,horizontal motor score,vertical motor score,1%sucrose water consumption rate,p-JAK1 and p-STAT3 protein relative expression levels in the Fructus Schisandrae Chinensis extracts groups were increased,and the apoptosis rate of hippocampal neurons was decreased(P<0.05),in which the changes of above indexes were most obvious in the high-dose Fructus Schisandrae Chinensis extracts group(P<0.05).Nissl’s staining results showed that hippocampal neurons in the model group were atrophied,disordered and sparsely arranged,with plenty necrotic cells,and compared with the model group,the above changes in various Fructus Schisandrae Chinensis extracts groups were improved in different degrees.Conclusion Fructus Schisandrae Chinensis extracts have significant anti-depressive effect,which may be related to the activation of JAK1/STAT3 signaling pathway to protect hippocampal neurons.
