摘要
目的研究黔产苗药土党参趁鲜蒸制炮制工艺,明确最佳炮制工艺条件;探讨土党参蒸制前后对脾虚模型小鼠的改善作用,进一步为临床应用提供参考。方法以党参炔苷、总多糖、醇溶性浸出物、水溶性浸出物为评价指标,采用综合评分法,优选土党参趁鲜蒸制炮制工艺;采用“大黄苦寒泻下+饮食失节+劳倦过度”联合法,建立小鼠脾虚动物模型,以参苓白术散为阳性对照,通过比较外观体征、脏器指数、负重游泳时间,检测谷丙转氨酶(ALT)、谷草转氨酶(AST)、碱性磷酸酶(ALP)、肌酐(CRE)、尿素氮(BUN)、干扰素(IFN-γ)、白介素4(IL-4)的含量,研究土党参趁鲜蒸制前后对小鼠脾虚动物模型的影响。结果不同炮制方法对土党参内在质量影响较大,趁鲜蒸制法综合评分值最高,其中趁鲜蒸制20 min(0.8672)>趁鲜蒸制50 min(0.7252)>趁鲜蒸制40 min(0.6846);而烘干后蒸制60 min(0.0025)综合评分值最低。同时,生品和趁鲜蒸制品均可改善小鼠脾虚动物模型外观体征(P<0.05),提高脾脏脏器指数(P<0.05),延长负重游泳时间(P<0.05),降低AST、ALT、ALP、CRE、BUN含量(P<0.01),减少IL-4/IFN-γ水平(P<0.01),且趁鲜蒸制品作用强于生品(P<0.05)。结论土党参优选的炮制方法为:取新鲜药材,洗净,趁鲜蒸制20 min后切厚片(2-4 mm),60℃烘干;趁鲜蒸制后对小鼠脾虚动物模型的改善作用较强。
Objective To study the processing technology of fresh steaming of Miao medicine Campanumoea javanica and determine the best processing conditions,and to further provide reference for clinical application based on the effect of steaming before and after on the animal model of spleen deficiency in mice.Methods Taking lobetyolin,total polysaccharide,alcohol-soluble extract and water-soluble extract as evaluation indexes,the processing technology of fresh steaming of Campanumoea javanica was optimized by comprehensive scoring method.The spleen deficiency animal model of mice was established by the combined method of"purgation by Rhei Radix et Rhizoma+diet disorder+overfatigue".Shenling Baizhu Powder was used as positive control,and the contents of alanine aminotransferase(ALT),aspartate aminotransferase(AST),alkaline phosphatase(ALP),creatinine(CRE),urea nitrogen(BUN),interferon(IFN-γ)and interleukin4(IL-4)were detected by comparing the physical signs,organ index and weight-bearing swimming time.To study the effect of Tudangshen on spleen deficiency animal model in mice before and after fresh steaming.Results Different processing methods had great influence on the internal quality of Campanumoea javanica,and the comprehensive score of fresh steaming method was the highest,with fresh steaming 20 min(0.8672)>fresh steaming 50 min(0.7252)>fresh steaming 40 min(0.6846).After drying for 60 min(0.0025),the comprehensive score was the lowest.Meanwhile,both raw and fresh steamed products could improve the appearance and physical signs of spleen deficiency in mouse model(P<0.05),increase spleen organ index(P<0.05),extend weight-bearing swimming time(P<0.05),and reduce the contents of AST,ALT,ALP,CRE and BUN(P<0.01).The level of IL-4/IFN-γwas decreased(P<0.01),and the effect of fresh steamed products was stronger than that of raw products(P<0.05).Conclusion The optimal processing method of Campanumoea javanica was as follows,taking fresh medicine and washing it,cutting thick slices(2-4 mm)after steaming for 20 min and drying at 60℃.Fresh steaming can improve the spleen deficiency model in mice.
