摘要
目的:建立高效液相色谱-蒸发光散射检测(HPLC-ELSD)法测定王不留行中多糖的含量。方法:采用Alltech Prevail Carbohydrate ES色谱柱(4.6mm×250mm,5μm);乙腈-水(75∶25)为流动相;柱温30℃;流量1.0mL/min。结果:王不留行多糖中单糖组分在1.267~27.140μg范围内呈良好线性关系(R;=0.9946),精密度RSD为2.0%。测定王不留行药材中多糖含量的重复性为1.3%,样品溶液在配制后24h内稳定性较好,平均回收率为98.6%,RSD为2.4%(n=6)。结论:该方法方便快捷,重现性好,结果可靠,可用于测定王不留行中多糖的含量。
Objective:To establish HPLC-ELSD method for the determination of polysaccharides in Semen vaccaria.Methods:The analysis was performed on Alltech Prevail TM Carbohydrate ES column(4.6 mm×250 mm,5μm),mobile phase was acetonitrilewater,column temperature was maintained at 30℃,and flow rate was 0.1 mL/min.Results:The polysaccharides in Semen vaccaria showed a good relationship within the range of 1.267~27.140μg,precision RSDis 2.0%.The repeatability of the determination of thepolysaccharide content in Semen vaccaria was 1.3%,the sample solution has good stability within 24 hours after preparation,the average recovery rate was 98.6%,RSD was 2.4%(n=6).Conclusion:The method is convenient and fast.It has good reproducibility and reliable results.It can be used to determine the content of polysaccharides in Semen vaccaria.
