摘要
目的探讨移植脂肪间充质干细胞(adipose-derived stem cells,ADSC)转化神经元移植对颞叶癫疒间大鼠学习记忆的影响。方法SD大鼠按实验要求,随机分成4组:移植组(n=15,颞叶癫疒间大鼠造模后在海马植入ADSC转化神经元),假移植组(n=15,颞叶癫疒间大鼠造模后在海马注射生理盐水),NS组(n=10,实验大鼠不造模,仅在海马注射生理盐水)和ADSC对照组(n=10,实验大鼠不造模,仅在海马植入ADSC转化神经元)。移植前、移植后2周、移植后2个月,分别进行Morris水迷宫实验,比较大鼠学习记忆能力的变化。结果移植后2周及2个月,与假移植组相比,移植组大鼠寻找平台平均潜伏期均缩短(P<0.05、P<0.01);而与NS组及ADSC对照组相比,无显著性差异(均P>0.05)。移植后2周及2个月,与NS组比较,移植组和ADSC对照组大鼠穿越Ⅱ象限时间,均无显著性差异(均P>0.05);而与移植组比较,假移植组大鼠穿越Ⅱ象限时间明显缩短(P<0.01)。结论ADSC转化神经元移植能改善颞叶癫疒间大鼠的学习记忆能力,且无明显副作用,具体机制尚待进一步研究。
Objective To investigate the effects oftransplantation ofadipose-derived stem cells(ADSCs)transformed neurons on the learning and memory of temporal lobe epilepsy rats.Methods The SD rats were randomly divided into 4 groups according to the experimental requirements:transplantation group(n=15,ADSC-transformed neurons were implanted into the hippocampus after modeling of temporal lobe epilepsy rats),sham-transplantation group(n=15,temporal lobe epilepsy rats after modeling,saline was injected into the hippocampus),NS group(n=10,experimental rats were not modeled,saline was injected only in the hippocampus)and ADSC control group(n=10,experimental rats were not modeled,only ADSC-transformed neurons were implanted into the hippocampus).Before transplantation,2 weeks and 2 months after transplantation,Morris water maze tests were performed to compare the changes in learning and memory abilities of rats.Results After 2 weeks and 2 months of transplantation,compared with the sham-transplantation group,the average latency of finding platform in the transplantation group was shorter(P<0.05,P<0.01);compared with the NS group and the ADSC control group,there was no significant difference(both P>0.05).After 2 weeks and 2 months of transplantation,compared with the NS group,there was no significant difference in the time across quadrantⅡbetween the transplantation group and the ADSC control group(both P>0.05);compared with the transplantation group,the time across quadrantⅡof the rats in the sham-transplantation group was significantly shorter(P<0.01).Conclusions The ADSC-transformed neuron transplantation can improve the learning and memory ability of temporal lobe epilepsy rats without significant side effects.The specific mechanism needs further study.
