The results showed that the antioxidant activity of polysaccharides in yam bulbils was signifi- cantly enhanced with the increase of concentration; they showed a strong scaveng- ing ability against DPPH. and .OH, and the scavenging ability was dose dependent to some extent; the scavenging rates reached 91.15% and 89.06% respectively when the dose reached 4.0 mg/ml; the polysaccharides in yam bulbils significantly educed the blood glucose in model rice, and the hypoglycemic effect of large-dose polysaccharides was more obvious. The polysaccharides in yam bulbils has good antioxidant activity and hypoglycemic effect, which provides a new source for devel- opment of safe and natural food antioxidants and blood sugar-lowering agents.展开更多

Chemical and spectroscopic studies led to the structure determination as follows: the backbone chain consists of 1, 6-linked-beta -D Galp, 1,4-linked-X -D Galp, 1,4-linked-beta -D GalA and 1,2- or 1,4-linked-beta -L Rhap having branching points at position O-3 of some 1,6-linked-beta -D Galp residues (one among eight) and O-4 or O-2 of 1,2- or 1,4-linked-beta -L Rhap residues to terminal beta -D-galactopyranose. The side chains composed of terminal Galp, 1,6-linked-beta -D Galp, 1,4-linked beta -D Galp and 1,3,6-linked-beta -D Galp also linked at position O-3 of 1,6-linked-beta -D Galp residues in the backbone chain. beta -L-arabinofuranosyl and terminal beta -L-rhamnopyranosyl residues existed in the periphery of this polysaccharide linked to O-3 of 1,6-linked-beta -D Galp residues in the backbone chain and the side chains. Single factor experiments were conducted to examine the effects of extracting time,temperature,the solid-liquid ratio and pH value on the extraction yield of polysaccharide from pumpkin. [Method] The best enzyme ratio and extraction conditions for complex enzymes extraction were determined through orthogonal tests. Scavenging ·OH and O-2 activities of pumpkin polysaccharides were also investigated by salicylic acid and improved self-oxidation of o-pheno methods respectively. [Results] The results showed that the biggest extraction yield of polysaccharide from pumpkin can be got when adding 1% cellulose enzyme,1.5% pectinase,1.0% papain and Na2HPO4-citric acid buffer solution (pH was 4.6),and oscillating for 30 min under water at 40 ℃ with the solid-liquid ratio of 1:30. In addition,pumpkin polysaccharides had a strong activity of eliminating ·OH,but very weak activity to scavenge O-2. [Method] The effects of extraction temperature, extraction time, extraction times and material-to-liquid ratio on content of polysaccharides in S. sagittifo/ia were investi- gated. On the basis of single-factor tests, an orthogonal test was carried out to de- termine the optimal process parameters. In addition, the in-vitro antioxidant activity of S. sagittifo/ia polysaccharides was evaluated on the basis of determination of their scavenging activities against DPPH and hydroxyl free radicals and reducing power. [Result] The results showed that the optimized process parameters were as follows: material-to-liquid ratio at 1:40 (g/ml), extraction temperature at 90 ℃, extrac- tion time of 4 h, and extraction times of 3 times. Under this condition, the content of S. sagittifo/ia polysaccharides was 29.32%. For 1 mg/ml S. sagittifo/ia polysaccha- rides, the scavenging ratios of DPPH and hydroxyl free radicals were 70.62% and 35.82%, respectively, and in the determination of reducing power, the absorbance at 700 nm was 0.453 1. [Conclusion] It could be concluded that S. sagittifolia polysac- charides have strong antioxidant capacity to have free radicals scavenged in vitro.展开更多

And their antioxidative as well as glycosidase inhibitory activities ar...In the present study, ultrasonic extraction technique (UET) is used to improve the yield of polysaccharides from Lami- naria japonica (LJPs). And their antioxidative as well as glycosidase inhibitory activities are investigated. Box-Behnken design (BBD) combined with response surface methodology (RSM) is applied to optimize ultrasonic extraction for polysaccharides. The optimized conditions are obtained as extraction time at 54 min, ultrasonic power at 1050 W, extraction temperature at 80℃ and ratio of material to solvent at 1:50 (g mL-1). Under these optimal ultrasonic extraction conditions, an actual experimental yield (5.75% + 0.3%) is close to the predicted result (5.67%) with no significant difference (P〉0.05). Vitro antioxidative and glycosidase inhibitory activities tests indicate that the crude polysaccharides (LJP) and two major ethanol precipitated fractions (LJP1 and LJP2) are in a concentration-dependent manner. LJP2 (30%-60% ethanol precipitated polysaccharides) possesses the strongest α-glucosidase in- hibitory activity and moderate scavenging activity against hydroxyl radicals (66.09% ±2.19%, 3.0 mg mL-l). Also, the inhibitory activity against a-glucosidase (59.08% ± 3.79%, 5.0 mg mL-1) is close to that of acarbose (63.99% ± 3.27%, 5.0 mg mL-l). LJP 1 (30% ethanol precipitated polysaccharides) exhibits the strongest scavenging activity against hydroxyl radicals (99.80%q-0.00%, 3.0mg mL-1) and moderate a-glucosidase inhibitory activity (47.76%± 1.92%, 5.0 mgmL-1). LJP shows the most remarkable DPPH scav- enging activity (66.20%±0.11%, 5.0mgmL-1) but weakest a-glucosidase inhibitory activity (37.77%±1.30%, 5.0mgmL-1). How- ever, all these LJPs exert weak inhibitory effects against a-amylase. These results show that UET is an effective method for extract- ing bioactive polysaccharides from seaweed materials. LJP 1 and LJP2 can be developed as a potential ingredient in hypoglycemic agents or functional food for the management of diabetes. This study provides scientific evidence and advances in the preparation technology and a hypoglycemic activities evaluation method for seaweed polysaccharides, especially glycosidase inhibition in com- bination with an antioxidative activity evaluation method.展开更多

Holothuriafuscogliva is a big sea cucumber with a rich of bioactive polysaccharides. To investigate the bioactivities of the polysaccharides from sea cucumber H. fuscogliva, we prepared the sulfated polysaccharides (HfP) from sea cucumber H. fuscogliva using a protease hydrolysis method. Antioxidant activities of HfP were investigated, including hydroxyl radical scavenging activity and superoxide radical scavenging activity. And, the anticoagulant activities of HfP were studied, including the activated partial thromboplastin time (APTT), prothrombin time (PT) and thrombin time (TT). The average molecular weight was 1 867.1 Da, with a sulfate content of 20.7%. In addition, the molar ratio of monosaccharide composition of HfP was Man: Rha: Glc A: Glc: Gal: Xyl: Fuc=0.083 6: 0.437: 0.134: 0: 1.182: 0.748: 1. It had a strong antioxidant activity, the hydroxyl and superoxide radical scavenging activity ECs0 of HfP was 3.74 and 0.037 mg/mL, respectively. It also showed a good anticoagulant activity in our study. The APTT of HfP was much higher than that of heparin sodium, and the PT and TT of HfP was close to that ofheparin sodium at a low concentration. Therefore, HfP shows a good antioxidant and anticoagulant activity and it may become a potential candidate of the natural antioxidant and anticoagulant and will have a good application future in health product or medicine industry.展开更多

A refined polysaccharide, named EPJ, was separated into nine fractions by DEAE-52 cellulose chromatogr...Polysaccharides were isolated from Semen Euryales by water extraction, alcohol precipitation and deproteinization. A refined polysaccharide, named EPJ, was separated into nine fractions by DEAE-52 cellulose chromatography and one of them was separated into two fractions by Sephadex G-100 column. The molecular weight of EPJ was determined to 15 367 Da and the monosaccharide composition was mainly glucose and rhamnose with the molar ratio of 5.46:1. The FT-IR spectra showed carbohydrate-related absorbance of polysaccharide functional groups. In vitro assay showed EPJ could significantly scavenge DPPH radical, superoxide anion, hydrogen peroxide and exhibited a strong reducing power. In vivo assay indicated that EPJ was effective in a D-galactose induced aging mice model. Results showed EPJ significantly inhibited formation of MDA and increased the activities of SOD, CAT, GSH-Px in a dose-dependent manner in mice tissues. The findings obtained from this study indicated that polysaccharides from Semen Euryales could be explored for a wide prospect of benefit.展开更多

And, the anticoagulant activities of HfP were studied, including the activated partial thromboplastin time (APTT), prothrombin time (PT) and thrombin time (TT). The average molecular weight was 1 867.1 Da, with a sulfate content of 20.7%. In addition, the molar ratio of monosaccharide composition of HfP was Man: Rha: Glc A: Glc: Gal: Xyl: Fuc=0.083 6: 0.437: 0.134: 0: 1.182: 0.748: 1. It had a strong antioxidant activity, the hydroxyl and superoxide radical scavenging activity ECs0 of HfP was 3.74 and 0.037 mg/mL, respectively. It also showed a good anticoagulant activity in our study. The APTT of HfP was much higher than that of heparin sodium, and the PT and TT of HfP was close to that ofheparin sodium at a low concentration. Therefore, HfP shows a good antioxidant and anticoagulant activity and it may become a potential candidate of the natural antioxidant and anticoagulant and will have a good application future in health product or medicine industry.
文摘Polysaccharides of Tricholoma matsutake Sing. were extracted with boilingwater, followed by precipitating with ethanol. Two membranes of different molecular weight cut-off, namely, 30 kD and 10 kD were chosen with ultrafiltration. The antioxidant activity of the purification polysaccharides was evaluated by various antioxidant assay, including DPPH (l,l-diphenyl-2-picrylhydrazyl) radical scavenging, superoxide anion radical scavenging and hydrogen peroxide scavenging. The scavenging effect on hydroxyl radical and DPPH radical, which may be comparable to Vitamin C, while the scavenging effect on superoxide anion radicals was not obvious.

文摘Polysaccharides were isolated from Semen Euryales by water extraction, alcohol precipitation and deproteinization. A refined polysaccharide, named EPJ, was separated into nine fractions by DEAE-52 cellulose chromatography and one of them was separated into two fractions by Sephadex G-100 column. The molecular weight of EPJ was determined to 15 367 Da and the monosaccharide composition was mainly glucose and rhamnose with the molar ratio of 5.46:1. The FT-IR spectra showed carbohydrate-related absorbance of polysaccharide functional groups. In vitro assay showed EPJ could significantly scavenge DPPH radical, superoxide anion, hydrogen peroxide and exhibited a strong reducing power. In vivo assay indicated that EPJ was effective in a D-galactose induced aging mice model. Results showed EPJ significantly inhibited formation of MDA and increased the activities of SOD, CAT, GSH-Px in a dose-dependent manner in mice tissues. The findings obtained from this study indicated that polysaccharides from Semen Euryales could be explored for a wide prospect of benefit.
