Effects of monoglyceride blend on systemic and intestinal immune responses,and gut health of weaned pigs experimentally infected with a pathogenic Escherichia coli

Background Monoglycerides have emerged as a promising alternative to conventional practices due to their biolog-ical activities,including antimicrobial properties.However,few studies have assessed the efficacy of monoglyceride blend on weaned pigs and their impacts on performance,immune response,and gut health using a disease chal-lenge model.Therefore,this study aimed to investigate the effects of dietary monoglycerides of short-and medium-chain fatty acids on the immunity and gut health of weaned pigs experimentally infected with an enterotoxigenic Escherichia coli F18.Results Pigs supplemented with high-dose zinc oxide(ZNO)had greater(P<0.05)growth performance than other treatments,but no difference was observed in average daily feed intake between ZNO and monoglycerides groups during the post-challenge period.Pigs in ZNO and antibiotic groups had lower(P<0.05)severity of diarrhea than control,but the severity of diarrhea was not different between antibiotic and monoglycerides groups.Pigs fed with monoglycerides or ZNO had lower(P<0.05)serum haptoglobin on d 2 or 5 post-inoculation than control.Pigs in ZNO had greater(P<0.05)goblet cell numbers per villus,villus area and height,and villus height:crypt depth ratio(VH:CD)in duodenum on d 5 post-inoculation than pigs in other treatments.Pigs supplemented with monoglyc-erides,ZNO,or antibiotics had reduced(P<0.05)ileal crypt depth compared with control on d 5 post-inoculation,contributing to the increase(P=0.06)in VH:CD.Consistently,pigs in ZNO expressed the lowest(P<0.05)TNFa,IL6,IL10,IL12,IL1A,IL1B,and PTGS2 in ileal mucosa on d 5 post-inoculation,and no difference was observed in the expres-sion of those genes between ZNO and monoglycerides.Supplementation of ZNO and antibiotic had significant impacts on metabolic pathways in the serum compared with control,particularly on carbohydrate and amino acid metabolism,while limited impacts on serum metabolites were observed in monoglycerides group when compared with control.Conclusions The results suggest that supplementation of monoglyceride blend may enhance disease resist-ance of weaned pigs by alleviating the severity of diarrhea and mitigating intestinal and systemic inflammation,although the effectiveness may not be comparable to high-dose zinc oxide. 展开更多

Background Enterotoxigenic Escherichia coli(E.coli)is a threat to humans and animals that causes intestinal dis-orders.Antimicrobial resistance has urged alternatives,including Lactobacillus postbiotics,to mitigate the effects of enterotoxigenic E.coli.Methods Forty-eight newly weaned pigs were allotted to NC:no challenge/no supplement;PC:F18^(+)E.coli chal-lenge/no supplement;ATB:F18^(+)E.coli challenge/bacitracin;and LPB:F18^(+)E.coli challenge/postbiotics and fed diets for 28 d.On d 7,pigs were orally inoculated withF18^(+)E.coli.At d 28,the mucosa-associated microbiota,immune and oxidative stress status,intestinal morphology,the gene expression of pattern recognition receptors(PRR),and intestinal barrier function were measured.Data were analyzed using the MIXED procedure in SAS 9.4.Results PC increased(P<0.05)Helicobacter mastomyrinus whereas reduced(P<0.05)Prevotella copri and P.ster-corea compared to NC.The LPB increased(P<0.05)P.stercorea and Dialister succinatiphilus compared with PC.The ATB increased(P<0.05)Propionibacterium acnes,Corynebacterium glutamicum,and Sphingomonas pseudosanguinis compared to PC.The PC tended to reduce(P=0.054)PGLYRP4 and increased(P<0.05)TLR4,CD14,MDA,and crypt cell proliferation compared with NC.The ATB reduced(P<0.05)NOD1 compared with PC.The LPB increased(P<0.05)PGLYRP4,and interferon-γand reduced(P<0.05)NOD1 compared with PC.The ATB and LPB reduced(P<0.05)TNF-αand MDA compared with PC.Conclusions TheF18^(+)E.coli challenge compromised intestinal health.Bacitracin increased beneficial bacteria show-ing a trend towards increasing the intestinal barrier function,possibly by reducing the expression of PRR genes.Lac-tobacillus postbiotics enhanced the immunocompetence of nursery pigs by increasing the expression of interferon-γand PGLYRP4,and by reducing TLR4,NOD1,and CD14. 展开更多

Enterotoxigenic E.coli is one of the bacterial pathogens contributing to the global resistance crisis in public health and animal husbandry.The problem of antibiotic resistance is becoming more and more serious,and phage is con-sidered one of the potential alternatives to antibiotics that could be utilized to treat bacterial infections.Our study isolated and identified a lytic phage PGX1 against multidrug-resistant enterotoxigenic E.coli EC6 strain from sew-age.The phage lysis profile revealed that PGX1 exhibited a lytic effect on multidrug-resistant enterotoxigenic E.coli strains of serotype O60.Through phage whole genome sequencing and bioinformatics analysis,PGX1 was found to be the class Caudoviricetes,family Autographiviridae,genus Teseptimavirus.The length of the PGX1 genome is about 37,009 bp,containing 54 open reading frames(ORFs).Notably,phage PGX1 lacks any lysogenic-related genes or virulence genes.Furthermore,phage PGX1 demonstrates strong adaptability,tolerance,and stability in various pH(pH4-10)and temperatures(4–40°C).The in vivo and in vitro tests demonstrated that phage PGX1 significantly removes and inhibits the formation of multidrug-resistant EC6 biofilm and effectively controls the Galleria mel-lonella larvae and enterotoxigenic E.coli EC6 during mice infection.In conclusion,the above findings demonstrated that phage PGX1 may be a novel antimicrobial agent to control multidrug-resistant E.coli infections. 展开更多

Escherichia coli O157:H7 is one of the major foodborne pathogenic bacterial that cause infectious diseases in humans.The previous found that a combination of kojic acid and tea polyphenols exhibited better activity against E.coli O157:H7 than using either alone.This study aimed to explore responses underlying the antibacterial mechanisms of kojic acid and tea polyphenols from the gene level.The functional enrichment analysis by comparing kojic acid and tea polyphenols individually or synergistically against E.coli O157:H7 found that acid resistance systems in kojic acid were activated,and the cell membrane and genomic DNA were destructed in the cells,resulting in“oxygen starvation”.The oxidative stress response triggered by tea polyphenols inhibited both sulfur uptake and the synthesis of ATP,which affected the bacteria's life metabolic process.Interestingly,we found that kojic acid combined with tea polyphenols hindered the uptake of iron that played an essential role in the synthesis of DNA,respiration,tricarboxylic acid cycle.The results suggested that the iron uptake pathways may represent a novel approach for kojic acid and tea polyphenols synergistically against E.coli O157:H7 and provided a theoretical basis for bacterial pathogen control in the food industry. 展开更多

BACKGROUND The association between the intestinal microbiota and psychiatric disorders is becoming increasingly apparent.The gut microbiota contributes to colorectal carcinogenesis(CRC),as demonstrated with colibactin-producing Escherichia coli(CoPEC).AIM To evaluate the association between CoPEC prevalence and anxiety-and depressive-like behaviors with both preclinical and clinical approaches.METHODS Patients followed after a CRC surgery and for whom the prevalence of CoPEC has been investigated underwent a psychiatric interview.Results were compared according to the CoPEC colonization.In parallel C57BL6/J wild type mice and mice with a CRC susceptibility were chronically infected with a CoPEC strain.Their behavior was assessed using the Elevated Plus Maze test,the Forced Swimming Test and the Behavior recognition system PhenoTyper®.RESULTS In a limited cohort,all patients with CoPEC colonization presented with psychiatric disorders several years before cancer diagnosis,whereas only one patient(17%)without CoPEC did.This result was confirmed in C57BL6/J wildtype mice and in a CRC susceptibility mouse model(adenomatous polyposis colimultiple intestinal neoplasia/+).Mice exhibited a significant increase in anxiety-and depressive-like behaviors after chronic infection with a CoPEC strain.CONCLUSION This finding provides the first evidence that CoPEC infection can induce microbiota-gut-brain axis disturbances in addition to its procarcinogenic properties. 展开更多

Avian pathogenic Escherichia coli(APEC)belonging to extraintestinal pathogenic E.coli(ExPEC)can cause severe infections in extraintestinal tissues in birds and humans,such as the lungs and blood.MprA(microcin production regulation,locus A,herein renamed AbsR,a blood survival regulator),a member of the MarR(multiple antibiotic resistance regulator)transcriptional regulator family,governs the expression of capsule biosynthetic genes in human ExPEC and represents a promising druggable target for antimicrobials.However,a deep understanding of the AbsR regulatory mechanism as well as its regulon is lacking.In this study,we present a systems-level analysis of the APEC AbsR regulon using ChIP-Seq(chromatin immunoprecipitation sequencing)and RNA-Seq(RNA sequencing)methods.We found that AbsR directly regulates 99 genes and indirectly regulates 667 genes.Furthermore,we showed that:1)AbsR contributes to antiphagocytotic effects by macrophages and virulence in a mouse model for systemic infection by directly activating the capsular gene cluster;2)AbsR positively impacts biofilm formation via direct regulation of the T2SS(type II secretion system)but plays a marginal role in virulence;and 3)AbsR directly upregulates the acid tolerance signaling system EvgAS to withstand acid stress but is dispensable in ExPEC virulence.Finally,our data indicate that the role of AbsR in virulence gene regulation is relatively conserved in ExPEC strains.Altogether,this study provides a comprehensive analysis of the AbsR regulon and regulatory mechanism,and our data suggest that AbsR likely influences virulence primarily through the control of capsule production.Interestingly,we found that AbsR severely represses the expression of the type I-F CRISPR(clustered regularly interspaced short palindromic repeats)-Cas(CRISPR associated)systems,which could have implications in CRISPR biology and application. 展开更多

Better understanding of their main resistance mechanisms to commonly used antibacterial agents will help to reduce the burden of this infection. The prevalence of drug resistant uropathogenic Escherichia coli isolates from immunocompromised diabetic patients attending selected health facilities in Benue State was investigated. Two hundred and ninety-six midstream urine samples were collected for both study and control diabetic patients. Bacterial isolation was done using semi-quantitative method. Drug resistant Escherichia coli were identified as multidrug resistant (MDR), extensive drug resistant (XDR) and pan-drug resistant organisms (PDR). Statistical significance was considered at p E. coli isolates from the study and control subjects with overall prevalence of 20.9% and 8.4% respectively. The isolates were highly resistant to penicillin (ampicillin), monobactam (aztreonam), older quinolone (nalidixic acid) whereas the majority of them showed high susceptibility to aminoglycoside (streptomycin), cephalosporin (cefotaxime) and carbapenem (imipenem). None showed complete susceptibility to all the tested antibiotics. Twenty-five E. coli were identified in this MDR, eight, XDR while 5 were PDR. High numbers of drug resistant E. coli isolates were identified in the study group of which 25 were MDR, 8 XDR while 5 were PDR isolates. High prevalence of UTI and drug resistant isolates occur in diabetic patients with hyperglycemic condition. 展开更多

Drug pressure is a factor that favors the emergence of a population of better adapted bacteria. However, there is no literature highlighting the genetic diversity and evolutionary structure of E. coli and K. pneumoniae in an environment with high selection pressure in Côte d’Ivoire. The objective of this study was to evaluate the genetic diversity of E. coli and K. pneumoniae strains circulating at the HKB Hospital in Abobo and at the Daloa Regional Hospital and its impact on the dissemination of extended spectrum beta-lactam resistance genes. A total of 39 strains isolated from the urinary tract of infected patients, including 30 strains of E. coli and 9 strains of K. pneumoniae were studied. A total of 39 strains isolated from the urinary tract of infected patients, including 30 strains of E. coli and 9 strains of K. pneumoniae were studied. From genomic DNA extracts, ESBL resistance genes were amplified by PCR and sequenced, in addition to genetic typing by ERIC-PCR. The data obtained were submitted to genetic and bioinformatics analyses. The results have shown a genetic diversity important in E. coli and K. pneumoniae with diversity indexs (SID) ranging from 0.5 to 0.77. The genetic structure of the bacterial species studied has shown a clonal distribution of strains with clones expressing TEM-9 and CTX-M-15 variants. Also, this clonal structure was correlated with the spread of resistance genes in E. coli and K. pneumoniae. The spread of resistant clones is a factor that might limit the fight against antibiotic resistance. 展开更多

Urinary tract infections (UTIs) are common infections caused by normal skin or rectum bacteria that get into the urethra and infect the urinary tract. Although the infection can affect various parts of the tract, bladder infections are the most prevalent kind. Uropathogenic Escherichia Coli (UPEC) is the most common pathogen associated with UTI development. Therefore, inhibiting the UPEC protein target (PDB ID: 8BVD) appears to be a promising therapeutic strategy. Therefore, in this study, molecular docking and dynamics were conducted to examine the antibacterial activity of Aloe barbadensis miller against UPEC bacteria. The Aloe barbadensis miller natural compounds licochalcone A, palmidin B and palmidin C were downloaded from PubChem with amoxicillin, which was used as a control drug and studied against the target molecule. The potential parameters examined were the docking scores, absorption, distribution, metabolism, excretion, toxicity (ADMET), bioavailability, root mean square deviation (RMSD), root mean square fluctuation (RMSF), hydrogen bonding, radius of gyration, and potential energy of the system. Docking scores showed that all ligands demonstrated an admirable candidature as an inhibitor to 8BVD molecule, and the score hierarchy is licochalcone A (-6.4 kcal/mol), palmidin C (-6.1 kcal/mol), palmidin B (-6.0 kcal/mol), and amoxicillin (-5.9 kcal/mol). All ligands appeared to have good drug-like properties and oral bioavailability. Molecular dynamic studies showed that all ligands exhibited an excellent nominee as inhibitors in their vicinity at 20 ns. However, there is a relatively high fluctuation of palmidin B compared with other compounds, which seems to be more stable. This work suggests that the selected phytoconstituents could be used as inhibitors of the 8BVD protein in the fight against UTIs. However, further investigation on the clinical and experimental validation of UTI treatment’s specific mechanisms and effects is still welcomed. 展开更多

Resistant bacteria can be transmitted to humans through feces or contaminated meat from local chickens. Bacterial strains were isolated from the intestinal contents of 400 local chicken samples from various sales sites. These strains were then characterized using bacteriological and biochemical methods to identify resistant strains. In a study conducted in Ouagadougou, we systematically collected chicken fecal samples from 20 locations across the city, followed by isolation and identification of Salmonella spp. using specific enrichment and culture methods, as well as Escherichia coli. Bacterial strains were characterized using antibiotic resistance profiles were determined through agar diffusion tests, revealing sensitivity or resistance to a range of antibiotics based on established scientific criteria. The results showed that out of the 400 samples collected, 81.25% and 63.5% were contaminated by Escherichia coli and Salmonella spp., respectively. Among these, 86.15% of identified Escherichia coli and 50.78% of Salmonella spp. displayed resistance to at least one tested antibiotic. Among 280 Escherichia coli isolates identified resistant to at least one antibiotic, 31.07% were resistant to cefotaxime (CTX), 20.35% to ceftazidime (CAZ), 21.07% to ceftriaxone (CTR), 75% to amoxicillin clavulanic acid (AMC), 23.57% aztreoname (ATM) and 27.14% were resistant to imipenem (IMP). In the case of the 129 Salmonella spp. isolates resistant to at least one tested antibiotic, 34.88% were resistant to CTX;41.08% to CAZ;35.65% to CTR, 92% to AMC, 39.53% to ATM and finally 47.28% were resistant to IMP. Our study revealed high prevalence of resistance in bacterial strains isolated from local chickens sold outdoors in Ouagadougou. These findings raise significant public health concerns, due to the possible transmission of these resistant strains to humans through the consumption of contaminated meat, thus complicating the treatment of bacterial infections. 展开更多

The aim of the present work was to evaluate the influence of Escherichia coli and Aeromonas hydrophila on some abiotic properties of groundwater stored under aerobic or anaerobic conditions. Experiments were performed in the presence of monospecific cells and bispecific cells. The incubation temperatures were 4˚C and 23˚C. The incubation (storage) times were 6 h, 12 h, 24 h, 48 h and 72 h. Bacteriological analyses were carried out using culture and physicochemical analyses using appropriate techniques. The results show that in the presence of monospecific cells and under aerobic conditions, E. coli abundances were relatively lower than under anaerobic conditions. The opposite was observed in the presence of bispecific cells. In most cases, minimum cell abundances were observed after 6 hours of storage. The same was true for A. hydrophila cells. The values of pH, electrical conductivity and ammonium ions in the water on the day of sampling varied over time. The pH values were relatively higher at 4˚C in the presence of E. coli cells alone, and at 23˚C in the presence of A. hydrophila cells. Ammonium ion levels were relatively higher at 4˚C under anaerobic conditions for both bacterial species. The highest electrical conductivity value was observed at 23˚C in the presence of E. coli cells, and at 4˚C in the presence of A. hydrophila cells, under anaerobic conditions. Electrical conductivity is higher in the presence of A. hydrophila with fluctuations from 169 to 285 µS/cm after 24 h incubation and 259 µS/cm after 6 hours incubation at 4˚C and 23˚C respectively. Under anaerobic conditions and in the presence of E. coli, the conductivity varied from 169 µS/cm at sampling to 274 µS/cm after 72 hours of incubation at 4˚C and from 169 to 289 after 24 hours. Under the same experimental conditions and in the presence of A. hydrophila, the electrical conductivity is higher at 4˚C with a value of 299 µS/cm after 24 hours. In the monospecific condition and under aerobic and anaerobic conditions and at an incubation temperature of 4˚C, the increase in the abundance of E. coli cells is significantly and positively correlated with the increase in pH values. In the mixed condition, pH is very significantly and positively correlated with the abundance of commensal Escherichia coli in anaerobic conditions (P NH4+) is significantly and negatively correlated with cellular concentrations of A. hydrophila in both aerobic and anaerobic conditions. 展开更多

The Transport and Persistence of Escherichia coli in Leachate from Poultry Litter Amended Soils

Fecal coliform bacteria such as Escherichia coli (E. coli) are one of the main sources of groundwater pollution. An assessment of the transport and Persistence of E. coli in poultry litter amended Decatur silty Clay soil and Hartsells Sandy soil was conducted using soil columns and simulated groundwater leaching. Enumeration of initial E. coli was determined to range from 2.851 × 10<sup>3</sup> to 3.044 × 10<sup>3</sup> CFU per gram of soil. These results have been used in a batch study to determine the persistence rate of E. coli in Decatur silty Clay soil and Hartsells Sandy soil. Results prove that E. coli survival growth rate increases for clay soil later than and at a higher rate than sandy soil. The column study has determined that E. coli was transported at a rate of 3.7 × 10<sup>6</sup><sup> </sup>CFU for Decatur silty loam and 6.3 × 10<sup>6</sup><sup> </sup>CFU for Hartsells sandy per gram of soil. Further, linear regression analysis predictions show higher porosity and soil moisture content affect transport, and Hartsells sandy soil has higher transport of E. coli due to its higher porosity and lower volumetric water content. 展开更多

This study tested that claim by comparing manuka honey used in medicine today, local unprocessed honey taken straight from a hive, and pasteurized honey found at a store, on strains of E. coli and S. epidermidis. The study evaluated the effects these honeys had on bacterial growth to determine which had the greatest inhibition of bacterial growth. To determine this, plates streaked with strains of E. coli or S. epidermidis bacteria and agar wells filled with one of the honeys were incubated and subsequently the diameter of the zone of inhibition was measured. After 20 trials using each honey and bacteria type, manuka and unprocessed were shown to have a statistically significant advantage over the pasteurized honey at inhibiting the growth of E. coli and S. epidermidis, though it was variable whether manuka had an advantage over the unprocessed honey. 展开更多

Lignocellulose pretreated using pyrolysis can yield clean energy(such as bioethanol)via microbial fermentation,which can significantly contribute to waste recycling,environmental protection,and energy security.However,the acids,aldehydes,and phenols present in bio-oil with inhibitory effects on microorganisms compromise the downstream utilization and conversion of lignocellulosic pyrolysates.In this study,we constructed a microbial electrolysis cell system for bio-oil detoxification and efficient ethanol production using evolved Escherichia coli to overcome the bioethanol production and utilization challenges highlighted in previous studies.In electrically treated bio-oil media,the E.coli-H strain exhibited significantly higher levoglucosan consumption and ethanol production capacities compared with the control.In undetoxified bio-oil media containing 1.0%(w/v)levoglucosan,E.coli-H produced 0.54 g ethanol/g levoglucosan,reaching 94%of the theoretical yield.Our findings will contribute to developing a practical method for bioethanol production from lignocellulosic substrates,and provide a scientific basis and technical demonstration for its industrialized application. 展开更多

Escherichia coli MLB(MG1655ΔpflBΔldhA),which can hardly grow on glucose with little succinate accumulation under anaerobic conditions.Two-stage fermentation is a fermentation in which the first stage is used for cell growth and the second stage is used for product production.The ability of glucose consumption and succinate production of MLB under anaerobic conditions can be improved significantly by using acetate as the solo carbon source under aerobic condition during the two-stage fermentation.Then,the adaptive laboratory evolution(ALE)of growing on acetate was applied here.We assumed that the activities of succinate production related enzymes might be further improved in this study.E.coli MLB46-05 evolved from MLB and it had an improved growth phenotype on acetate.Interestingly,in MLB46-05,the yield and tolerance of succinic acid in the anaerobic condition of two-stage fermentation were improved significantly.According to transcriptome analysis,upregulation of the glyoxylate cycle and the activity of stress regulatory factors are the possible reasons for the elevated yield.And the increased tolerance to acetate made it more tolerant to high concentrations of glucose and succinate.Finally,strain MLB46-05 produced 111 g/L of succinic acid with a product yield of 0.74 g/g glucose. 展开更多

Biosynthesis of paclitaxel(Taxol™)is a hot topic with extensive and durable interests for decades.However,it is severely hindered due to the very low titers of intermediates.In this study,Escherichia coli was employed to de novo synthesize a key intermediate of paclitaxel,taxadien-5α-yl-acetate(T5OAc).Plasmid-based pathway reconstruction and optimization were conducted for T5OAc production.The endogenous methylerythritol phosphate pathway was enhanced to increase the precursor supply.Three taxadien-5α-ol O-acetyltransferases were tested to obtain the best enzyme for the acetylation step.Metabolic burden was relieved to restore cell growth and promote production through optimizing the plasmid production system.In order to achieve metabolic balance,the biosynthesis pathway was regulated precisely by multivariate-modular metabolic engineering.Finally,in a 5-L bioreactor,the T5OAc titer was enhanced to reach 10.9 mg/L.This represents an approximately 272-fold increase in production compared to the original strain,marking the highest yield of T5OAc ever documented in E.coli,which is believed to be helpful for promoting the progress of paclitaxel biosynthesis. 展开更多

[Objective] The aim of this study was to explore the sterilization effects on Escherichia coli by adding bacterial inhibitor(CaCN2)during the process of cattle manure composting so as to provide a theoretical basis for cattle manure harmless treatment.[Method] Both experimental groups supplemented with 2.0% bacterial inhibitor and control groups without bacterial inhibitor were cultured under different temperatures(20,30,37,50,60 ℃)to determine the optimal composing temperature.Under 30 ℃,different bacterial inhibitor doses(0,2.0%,2.5%,3.0%)were added into the compost to obtain the optimal bacterial inhibitor addition dose.[Result] 30,50 and 60 ℃ were ideal temperatures for sterilization of E.coli.Under 30 ℃,E.coli couldn't be detected in 2.5% dose group and 3.0% dose group after culture for 48 h,demonstrating no less than 2.5% bacterial inhibitor should be added.[Conclusion] It has an important significance to enhance the sterilization effects on E.coli by adding CaCN2 into cattle manure compost especially in winter. 展开更多
