BACKGROUND The annual incidence of metabolic-associated fatty liver disease(MAFLD)in China has been increasing and is often overlooked owing to its insidious charac-teristics.Approximately 50%of the patients have a no...BACKGROUND The annual incidence of metabolic-associated fatty liver disease(MAFLD)in China has been increasing and is often overlooked owing to its insidious charac-teristics.Approximately 50%of the patients have a normal weight or are not obese.They are said to have lean-type MAFLD,and few studies of such patients are available.Because MAFLD is associated with abnormal lipid metabolism,lipid-targeted metabolomics was used in this study to provide experimental evidence for early diagnosis and pathogenesis.MAFLD and analyze metabolic pathways.UPLC-Q-Orbitrap/MS content determination was used to determine serum palmitic acid(PA),oleic acid(OA),linoleic acid(LA),and arachidonic acid(AA)levels in lean-type MAFLD patients.RESULTS Urea nitrogen and uric acid levels were higher in lean-type MAFLD patients than in healthy individuals(P<0.05).Alanine transaminase and cholinesterase levels were higher in lean-type MAFLD patients than in healthy indi-viduals(P<0.01).The expression of high-density lipoprotein and apolipoprotein A-1 were lower in lean-type MAFLD patients than in healthy individuals(P<0.05)and the expression of triglycerides and fasting blood glucose were increased(P<0.01).A total of 65 biomarkers that affected the synthesis and metabolism of fatty acids were found with P<0.05 and variable importance in projection>1.The levels of PA,OA,LA,and AA were significantly increased compared with healthy individuals.CONCLUSION The metabolic profiles of lean-type MAFLD patients and healthy participants differed significantly,yielding 65 identified biomarkers.PA,OA,LA,and AA exhibited the most significant changes,offering valuable clinical guidance for prevention and treatment of lean-type MAFLD.展开更多

When voltage flicker or...Loads in large scale industrial enterprises are always various and complex, causing insecure power quality. Meanwhile, some sensitive loads or precise instruments demand for high power quality. When voltage flicker or harmonics occurs, dynamic voltage restorer (DVR) can be used to stabilize the load-side voltage, which is a good manner of improving power quality. The paper explains the load demand in large scale industrial enterprises, analyzes several common voltage compensation scheme of DVR, and comes up with another voltage compensation scheme of DVR applied for double power supply with double load. A Simulink model according to the new scheme is also built in MATLAB/Simulink to test its performance. Simulation results show that the proposed scheme can guarantee the load-side voltage stability, which is an effective scheme for managing power quality.展开更多

METHODS: Forty-five male Sprague-Dawley rats we...AIM: To investigate the effect of glycyrrhizic acid (GA) on carbon tetrachloride (CCl4)-induced hepatocyte apo-ptosis in rats via a p53-dependent mitochondrial path-way. METHODS: Forty-five male Sprague-Dawley rats were randomly and equally divided into three groups, the control group, the CCl4 group, and the GA treatment group. To induce liver fibrosis in this model, rats were given a subcutaneous injection of a 40% solution of CCl4 in olive oil at a dose of 0.3 mL/100 g body weight biweekly for 8 wk, while controls received the same isovolumetric dose of olive oil by hypodermic injection, with an initial double-dose injection. In the GA group,rats were also treated with a 40% solution of CCl4 plus 0.2% GA solution in double distilled water by the intraperitoneal injection of 3 mL per rat three times a week from the first week following previously published methods, with modifications. Controls were given the same isovolumetric dose of double distilled water. Liver function parameters, such as alanine aminotransferase (ALT) and aspartate aminotransferase (AST) were de-termined. Pathologic changes in the liver were detected by hematoxylin and eosin staining. Collagen fibers were evaluated by Sirius red staining. Hepatocyte apoptosis was investigated using the terminal deoxynucleotidyl transferase-mediated deoxyuridine 5-triphosphate nick end labeling (TUNEL) assay and the cleaved caspase-3 immunohistochemistry assay. The expression levels of p53 and apoptosis-related proteins were evaluated by immunohistochemistry or Western blotting analysis. RESULTS: After 8 wk of treatment, GA significantly re-duced serum activity of ALT (from 526.7 ± 57.2 to 342 ± 44.8, P<0.05) and AST (from 640 ± 33.7 to 462.8 ± 30.6, P<0.05), attenuated the changes in liver his-topathology and reduced the staging score (from 3.53 ± 0.74 to 3.00 ± 0.76, P<0.05) in CCl4 -treated rats. GA markedly reduced the positive area of Sirius red and the ratio of the hepatic fibrotic region (from 7.87% ± 0.66% to 3.68% ± 0.32%, P<0.05) compared with the CCl4 group. GA also decreased the expression level of cleaved caspase-3 compared to the CCl4 group. TU-NEL assay indicated that GA significantly diminished the number of TUNEL-positive cells compared with the CCl4 group (P<0.05). GA treatment clearly decreased the level of p53 (P<0.05) detected by immunohis-tochemistry and Western blotting analysis. Compared with the CCl4 group, we also found that GA reduced the Bax/Bcl-2 ratio (P<0.05), the expression of cleaved caspase-3 (P<0.05), cleaved caspase-9 (P<0.05), and inhibited cytochrome C and second mitochondria-derived activator of caspases (Smac) release from mito-chondria to cytoplasm, i.e. , GA reduced the expressionlevel of Smac, which inhibited c-IAP1 activity (P<0.05), ultimately inhibiting the activity of caspase-3, according to Western blotting analysis. BACKGROUND The recovery time of hand wounds is long,which can easily result in chronic and refractory wounds,making the wounds unable to be properly repaired.The treatment cycle is long,the cost is high,and it is pron...BACKGROUND The recovery time of hand wounds is long,which can easily result in chronic and refractory wounds,making the wounds unable to be properly repaired.The treatment cycle is long,the cost is high,and it is prone to recurrence and disability.Double layer artificial dermis combined with autologous skin transplantation has been used to repair hypertrophic scars,deep burn wounds,exposed bone and tendon wounds,and post tumor wounds.AIM To investigate the therapeutic efficacy of autologous skin graft transplantation in conjunction with double-layer artificial dermis in treating finger skin wounds that are chronically refractory and soft tissue defects that expose bone and tendon.METHODS Sixty-eight chronic refractory patients with finger skin and soft tissue defects accompanied by bone and tendon exposure who were admitted from July 2021 to June 2022 were included in this study.The observation group was treated with double layer artificial dermis combined with autologous skin graft transplantation(n=49),while the control group was treated with pedicle skin flap transplantation(n=17).The treatment status of the two groups of patients was compared,including the time between surgeries and hospital stay.The survival rate of skin grafts/flaps and postoperative wound infections were evaluated using the Vancouver Scar Scale(VSS)for scar scoring at 6 mo after surgery,as well as the sensory injury grading method and two-point resolution test to assess the recovery of skin sensation at 6 mo.The satisfaction of the two groups of patients was also compared.RESULTS Wound healing time in the observation group was significantly longer than that in the control group(P<0.05,27.92±3.25 d vs 19.68±6.91 d);there was no significant difference in the survival rate of skin grafts/flaps between the two patient groups(P>0.05,95.1±5.0 vs 96.3±5.6).The interval between two surgeries(20.0±4.3 d)and hospital stay(21.0±10.1 d)in the observation group were both significantly shorter than those in the control group(27.5±9.3 d)and(28.4±17.7 d),respectively(P<0.05).In comparison to postoperative infection(23.5%)and subcutaneous hematoma(11.8%)in the control group,these were considerably lower at(10.2%)and(6.1%)in the observation group.When comparing the two patient groups at six months post-surgery,the excellent and good rate of sensory recovery(91.8%)was significantly higher in the observation group than in the control group(76.5%)(P<0.05).There was also no statistically significant difference in two point resolution(P>0.05).The VSS score in the observation group(2.91±1.36)was significantly lower than that in the control group(5.96±1.51),and group satisfaction was significantly higher(P<0.05,90.1±6.3 vs 76.3±5.2).CONCLUSION The combination of artificial dermis and autologous skin grafting for the treatment of hand tendon exposure wounds has a satisfactory therapeutic effect.It is a safe,effective,and easy to operate treatment method,which is worthy of clinical promotion.展开更多

Data deduplication removes duplicate chunks and becomes an important tech...An abundance of data have been generated from various embedded devices, applications, and systems, and require cost-efficient storage services. Data deduplication removes duplicate chunks and becomes an important technique for storage systems to improve space efficiency. However, stored unique chunks are heavily fragmented, decreasing restore performance and incurs high overheads for garbage collection. Existing schemes fail to achieve an efficient trade-off among deduplication, restore and garbage collection performance, due to failing to explore and exploit the physical locality of different chunks. In this paper, we trace the storage patterns of the fragmented chunks in backup systems, and propose a high-performance deduplication system, called HiDeStore. The main insight is to enhance the physical-locality for the new backup versions during the deduplication phase, which identifies and stores hot chunks in the active containers. The chunks not appearing in new backups become cold and are gathered together in the archival containers. Moreover, we remove the expired data with an isolated container deletion scheme, avoiding the high overheads for expired data detection. Compared with state-of-the-art schemes, HiDeStore improves the deduplication and restore performance by up to 1.4x and 1.6x, respectively, without decreasing the deduplication ratios and incurring high garbage collection overheads.展开更多
Polymyxin B(PB),as the last-line of defense against multidrug-resistant Gram-negative bacteria,has caused resistance to P.aeruginosa recently.Fortunately,synergistic treatment could preserve the last class of antibiot...Polymyxin B(PB),as the last-line of defense against multidrug-resistant Gram-negative bacteria,has caused resistance to P.aeruginosa recently.Fortunately,synergistic treatment could preserve the last class of antibiotics and reduce the emergency of drug resistance.Here,we performed a screen of 970 approved drugs synergized with PB against the P.aeruginosa DK2,which is severely resistant to PB,MIC=512μg/mL.Encouragingly,we found fluoroquinolones could synergy with PB and achieved an obvious reduction in MIC of PB below the clinical susceptible breakpoint(2 μg/mL).Especially,gemifloxacin achieved the highest synergistic effect with PB,leading to a 4096-fold MIC reduction(reduced from512 μg/mL to 0.125 μg/mL).Furthermore,synergistic effect was also observed in the combination of gemifloxacin and colistin.Finally,outer membrane permeabilization assay showed that gemifloxacin could increase the permeability of bacterial cell membranes for P.aeruginosa which partly explained the synergy mechanism.These results indicate that fluoroquinolones represent attractive synergists to address the emerging threat of polymyxin-resistant infections.展开更多
During infections,bacteria stimulate host cells to produce and release histamine,which is a key mediator of vital cellular processes in animals.However,the mechanisms underlying the bacterial cell’s ability to sense ...During infections,bacteria stimulate host cells to produce and release histamine,which is a key mediator of vital cellular processes in animals.However,the mechanisms underlying the bacterial cell’s ability to sense and respond to histamine are poorly understood.Herein,we show that HinK,a Lys R-type transcriptional regulator,is required to evoke responses to histamine in Pseudomonas aeruginosa,an important human pathogen.HinK directly binds to and activates the promoter of genes involved in histamine uptake and metabolism,iron acquisition,and Pseudomonas quinolone signal(PQS)biosynthesis.The transcriptional regulatory activity of HinK is induced when histamine is present,and it occurs when HinK binds with imidazole-4-acetic acid(Im AA),a histamine metabolite whose production in P.aeruginosa depends on the HinK-activated histamine uptake and utilization operon hin DAC-pa0222.Importantly,the inactivation of HinK inhibits diverse pathogenic phenotypes of P.aeruginosa.These results suggest that histamine acts as an interkingdom signal and provide insights into the mechanism used by pathogenic bacteria to exploit host regulatory signals to promote virulence.展开更多
A bacterium strain Y3,capable of efficiently degrading pendimethalin,was isolated from activated sludge and identified as Bacillus subtilis according to its phenotypic features and 16 S rRNA phylogenetic analysis.This...A bacterium strain Y3,capable of efficiently degrading pendimethalin,was isolated from activated sludge and identified as Bacillus subtilis according to its phenotypic features and 16 S rRNA phylogenetic analysis.This strain could grow on pendimethalin as a sole carbon source and degrade 99.5%of 100 mg/L pendimethalin within 2.5 days in batch liquid culture,demonstrating a greater efficiency than any other reported strains.Three metabolic products,6-aminopendimethalin,5-amino-2-methyl-3-nitroso-4-(pentan-3-ylamino) benzoic acid,and 8-amino-2-ethyl-5-(hydroxymethyl)-1,2-dihydroquinoxaline-6-carboxylic acid,were identified by HPLC-MS/MS,and a new microbial degradation pathway was proposed.A nitroreductase catalyzing nitroreduction of pendimethalin to 6-aminopendimethalin was detected in the cell lysate of strain Y3.The cofactor was nicotinamide adenine dinucleotide phosphate(NADPH) or more preferably nicotinamide adenine dinucleotide(NADH).The optimal temperature and pH for the nitroreductase were 30℃ and 7.5,respectively.Hg^(2+),Ni^(2+),Pb^(2+),Co^(2+),Mn^(2+) Cu^(2+),Ag~+,and EDTA severely inhibited the nitroreductase activity,whereas Fe^(2+),Mg^(2+),and Ca^(2+) enhanced it.This study provides an efficient pendimethalin-degrading microorganism and broadens the knowledge of the microbial degradation pathway of pendimethalin.展开更多

文摘BACKGROUND The recovery time of hand wounds is long,which can easily result in chronic and refractory wounds,making the wounds unable to be properly repaired.The treatment cycle is long,the cost is high,and it is prone to recurrence and disability.Double layer artificial dermis combined with autologous skin transplantation has been used to repair hypertrophic scars,deep burn wounds,exposed bone and tendon wounds,and post tumor wounds.AIM To investigate the therapeutic efficacy of autologous skin graft transplantation in conjunction with double-layer artificial dermis in treating finger skin wounds that are chronically refractory and soft tissue defects that expose bone and tendon.METHODS Sixty-eight chronic refractory patients with finger skin and soft tissue defects accompanied by bone and tendon exposure who were admitted from July 2021 to June 2022 were included in this study.The observation group was treated with double layer artificial dermis combined with autologous skin graft transplantation(n=49),while the control group was treated with pedicle skin flap transplantation(n=17).The treatment status of the two groups of patients was compared,including the time between surgeries and hospital stay.The survival rate of skin grafts/flaps and postoperative wound infections were evaluated using the Vancouver Scar Scale(VSS)for scar scoring at 6 mo after surgery,as well as the sensory injury grading method and two-point resolution test to assess the recovery of skin sensation at 6 mo.The satisfaction of the two groups of patients was also compared.RESULTS Wound healing time in the observation group was significantly longer than that in the control group(P<0.05,27.92±3.25 d vs 19.68±6.91 d);there was no significant difference in the survival rate of skin grafts/flaps between the two patient groups(P>0.05,95.1±5.0 vs 96.3±5.6).The interval between two surgeries(20.0±4.3 d)and hospital stay(21.0±10.1 d)in the observation group were both significantly shorter than those in the control group(27.5±9.3 d)and(28.4±17.7 d),respectively(P<0.05).In comparison to postoperative infection(23.5%)and subcutaneous hematoma(11.8%)in the control group,these were considerably lower at(10.2%)and(6.1%)in the observation group.When comparing the two patient groups at six months post-surgery,the excellent and good rate of sensory recovery(91.8%)was significantly higher in the observation group than in the control group(76.5%)(P<0.05).There was also no statistically significant difference in two point resolution(P>0.05).The VSS score in the observation group(2.91±1.36)was significantly lower than that in the control group(5.96±1.51),and group satisfaction was significantly higher(P<0.05,90.1±6.3 vs 76.3±5.2).CONCLUSION The combination of artificial dermis and autologous skin grafting for the treatment of hand tendon exposure wounds has a satisfactory therapeutic effect.It is a safe,effective,and easy to operate treatment method,which is worthy of clinical promotion.

文摘Objective:To explore the clinical significance of serum tumor markers and cytokines in the detection of breast cancer.Methods:A total of 586 different breast cancer staging, 250 patients with benign breast disease and 250 controls were detected and compared the serum tumor markers (CA125, CA153, CA199, CEA,β-hCG, CYFRA21-1, TPS) and cytokines (TNF-α, IL-6, IL-8, GDF3) level.Results:The serum tumor staging of breast cancer were different markers (CA125, CA153, CA199, CEA,β-hCG, CYFRA21-1, TPS) and cytokines (TNF-α, IL-6, IL-8, GDF3) were higher than those in control group and benign breast disease group and had statistical difference, serum tumor between different groups of breast cancer staging markers and cytokine levels were statistically different, with the staging of elevated serum tumor markers were increased with stage IV breast cancer group was the highest, was found between the control group and the benign breast disease group serum tumor marker was found the difference and cytokine.Conclusion:Serum tumor markers (CA125, CA153, CA199, CEA,β-hCG, CYFRA21-1, TPS) and cytokines (TNF-α, IL-6, IL-8, GDF3) in breast cancer detection have important clinical significance.

文摘An abundance of data have been generated from various embedded devices, applications, and systems, and require cost-efficient storage services. Data deduplication removes duplicate chunks and becomes an important technique for storage systems to improve space efficiency. However, stored unique chunks are heavily fragmented, decreasing restore performance and incurs high overheads for garbage collection. Existing schemes fail to achieve an efficient trade-off among deduplication, restore and garbage collection performance, due to failing to explore and exploit the physical locality of different chunks. In this paper, we trace the storage patterns of the fragmented chunks in backup systems, and propose a high-performance deduplication system, called HiDeStore. The main insight is to enhance the physical-locality for the new backup versions during the deduplication phase, which identifies and stores hot chunks in the active containers. The chunks not appearing in new backups become cold and are gathered together in the archival containers. Moreover, we remove the expired data with an isolated container deletion scheme, avoiding the high overheads for expired data detection. Compared with state-of-the-art schemes, HiDeStore improves the deduplication and restore performance by up to 1.4x and 1.6x, respectively, without decreasing the deduplication ratios and incurring high garbage collection overheads.

文摘Polymyxin B(PB),as the last-line of defense against multidrug-resistant Gram-negative bacteria,has caused resistance to P.aeruginosa recently.Fortunately,synergistic treatment could preserve the last class of antibiotics and reduce the emergency of drug resistance.Here,we performed a screen of 970 approved drugs synergized with PB against the P.aeruginosa DK2,which is severely resistant to PB,MIC=512μg/mL.Encouragingly,we found fluoroquinolones could synergy with PB and achieved an obvious reduction in MIC of PB below the clinical susceptible breakpoint(2 μg/mL).Especially,gemifloxacin achieved the highest synergistic effect with PB,leading to a 4096-fold MIC reduction(reduced from512 μg/mL to 0.125 μg/mL).Furthermore,synergistic effect was also observed in the combination of gemifloxacin and colistin.Finally,outer membrane permeabilization assay showed that gemifloxacin could increase the permeability of bacterial cell membranes for P.aeruginosa which partly explained the synergy mechanism.These results indicate that fluoroquinolones represent attractive synergists to address the emerging threat of polymyxin-resistant infections.

文摘During infections,bacteria stimulate host cells to produce and release histamine,which is a key mediator of vital cellular processes in animals.However,the mechanisms underlying the bacterial cell’s ability to sense and respond to histamine are poorly understood.Herein,we show that HinK,a Lys R-type transcriptional regulator,is required to evoke responses to histamine in Pseudomonas aeruginosa,an important human pathogen.HinK directly binds to and activates the promoter of genes involved in histamine uptake and metabolism,iron acquisition,and Pseudomonas quinolone signal(PQS)biosynthesis.The transcriptional regulatory activity of HinK is induced when histamine is present,and it occurs when HinK binds with imidazole-4-acetic acid(Im AA),a histamine metabolite whose production in P.aeruginosa depends on the HinK-activated histamine uptake and utilization operon hin DAC-pa0222.Importantly,the inactivation of HinK inhibits diverse pathogenic phenotypes of P.aeruginosa.These results suggest that histamine acts as an interkingdom signal and provide insights into the mechanism used by pathogenic bacteria to exploit host regulatory signals to promote virulence.

文摘A bacterium strain Y3,capable of efficiently degrading pendimethalin,was isolated from activated sludge and identified as Bacillus subtilis according to its phenotypic features and 16 S rRNA phylogenetic analysis.This strain could grow on pendimethalin as a sole carbon source and degrade 99.5%of 100 mg/L pendimethalin within 2.5 days in batch liquid culture,demonstrating a greater efficiency than any other reported strains.Three metabolic products,6-aminopendimethalin,5-amino-2-methyl-3-nitroso-4-(pentan-3-ylamino) benzoic acid,and 8-amino-2-ethyl-5-(hydroxymethyl)-1,2-dihydroquinoxaline-6-carboxylic acid,were identified by HPLC-MS/MS,and a new microbial degradation pathway was proposed.A nitroreductase catalyzing nitroreduction of pendimethalin to 6-aminopendimethalin was detected in the cell lysate of strain Y3.The cofactor was nicotinamide adenine dinucleotide phosphate(NADPH) or more preferably nicotinamide adenine dinucleotide(NADH).The optimal temperature and pH for the nitroreductase were 30℃ and 7.5,respectively.Hg^(2+),Ni^(2+),Pb^(2+),Co^(2+),Mn^(2+) Cu^(2+),Ag~+,and EDTA severely inhibited the nitroreductase activity,whereas Fe^(2+),Mg^(2+),and Ca^(2+) enhanced it.This study provides an efficient pendimethalin-degrading microorganism and broadens the knowledge of the microbial degradation pathway of pendimethalin.
文摘Synthetic biology,gene editing and transgene technologies that involving gene manipulation are widely used in food research and development.In recent decades,China has formulatedmany policies and regulations to govern the safety of genetically modified agricultural organisms.Based on the comparative analysis of technical connotations on synthetic biology,gene editing and transgenes and related risksregarding the safety of food,environment and the management of genetically modified agricultural organisms,this paper puts forward the following suggestions on 1)management of synthetic biological and gene-edited foods(in this paper,“food”refers to“edible agricultural products and food”),2)promoting the gradual process of industrialization on gene-edited and synthetic biological foods through lessons learned from the practice on genetically modified crops,3)speeding up the industrialization of gene-edited foods and prudently promoting the application of synthetic biological foods,4)urgently establishing and updating policies and regulations related to gene-edited and synthetic biological foods,5)unifying the legislation and supervision of synthetic biological,gene-edited and genetically modified foods 6)establishing ethical principles and bottom lines for genetically manipulated foods.
