Protective effects of new DNA vaccine for herpes simplex virus

摘要 目的 构建新型单纯疱疹病毒Ⅱ型DNA疫苗 ,鉴定其在哺乳细胞中的表达 ,检测其激发动物产生免疫反应的能力。方法 RT -PCR鉴定gD基因在COS - 7细胞中的表达 ,ELISA检测血清中中和抗体水平 ,用病毒攻击后检测其保护效率。结果 pVAX -gD能在COS- 7细胞中表达 ,并可激发免疫动物产生高滴度中和抗体 ,有效保护动物免受致死量病毒的攻击。 ObjectiveTo construct a new herpes simplex virus DNA vaccine, to identify its expression in mammalian cells, and to investigate its ability to induce immune response in animals. MethodsExpression of gD gene in COS-7 cell was detected with reverse transcription PCR. IgG in serum was asssayed with ELISA and protective effect of DNA vaccine was also investigated. ResultsThe constructed new DNA vaccine pVAX-gD can express the foreign gene gD in COS-7 cell, induce high titer of IgG in animals and protect the animals from lethal viral challenge. ConclusionThe constructed new DNA vaccine is a promising vaccine strain for human genital herpes.
