cDNA cloning of complete adiponectin gene coding sequence from Chinese

摘要 目的 克隆人脂联素 (ADPN)基因cDNA ,为进一步研究ADPN的功能提供实验基础。方法 应用RT PCR方法从中国人大网膜脂肪垫总RNA中扩增出ADPNcDNA全长基因 ,克隆入载体 pMD18 T中 ,形成重组载体pMD18 T/人ADPN。筛选出阳性克隆 ,通过限制性内切酶酶切鉴定后对其进行测序。结果 从脂肪组织总RNA中扩增得到74 5bp片段ADPN基因 ,其cDNA序列与GenBank人ADPN基因序列相同。结论 成功地克隆ADPN基因全长cDNA。 Objective To provide the experimental basis for further research on adiponectin (ADPN) function,human ADPN(hADPN) gene was cloned. Methods ADPN mRNA from greater omentum fat pat of Chinese was amplified by RT-PCR and cloned into plasmid pMD18-T to generate the recombinant plasmid pMD18-T/hADPN,which was confirmed to contain the amplified target gene segments by digest of restrictive endonuclease and nucleotide sequencing. Results The sequencing results for amplified target gene showed that the sequence of hADPN from omentum fat pat of Chinese was identical to that of hADPN in Genbank. Conclusion Complete ADPN cDNA is successfully cloned from Chinese.
