摘要
选取9种异硫氰酸酯类香料,研究其对常见的两种革兰氏阴性致病菌——副溶血性弧菌和沙门氏菌的抑菌活性。通过倍比肉汤稀释法测定最低抑菌浓度,根据OD600nm值绘制生长曲线。使用荧光定量PCR方法研究异硫氰酸酯类香料对副溶血性弧菌主要的毒力基因——tdh基因表达的影响。结果表明,调整香料浓度为0.04mol/L时,异硫氰酸苄酯、异硫氰酸3-甲硫基丙酯、异硫氰酸苯乙酯、异硫氰酸丁酯和异硫氰酸4-戊烯酯对两种供试菌均有明显的抑制作用(P<0.05)。其中异硫氰酸苄酯的抑菌效果最佳,对副溶血性弧菌和沙门氏菌的最低抑菌浓度分别为9.54μmol/L和2.44 mmol/L。异硫氰酸苄酯对tdh的基因表达具有抑制作用,并且有浓度依赖性。综上,异硫氰酸苄酯对两种革兰氏阴性致病菌有很好的作用效果,苄基对异硫氰酸酯类香料抑制革兰氏阴性致病菌有一定的影响,说明异硫氰酸苄酯香料可作为抑制致病菌的添加剂应用。
Nine kinds of isothiocyanate flavors were selected and their antibacterial activities against two common gram-negative bacteria including Vibrio parahaemolyticus and Salmonella were investigated. The minimum inhibitory concentration(MIC) was determined by broth dilution method, and the growth curve was obtained basing on OD600 nmvalues.The effects of isothiocyanate flavors on gene expression of major virulence gene tdh of V. parahaemolyticus were also studied by real-time PCR methods. The results showed that when the nine kinds of isothiocyanate flavors’ concentration was adjusted to 0.04 mol/L, benzyl isothiocyanate(BZITC), 3-methylthiopropyl isothiocyanate(MTPITC), phenethyl isothiocyanate(PEITC), butyl isothiocyanate(BUITC) and 4-pentenyl isothiocyanate(PTITC) had significant inhibitory effects against two tested bacteria(P <0.05). BZITC showed the strongest inhibitory effect with the MIC values of 9.54 μmol/L and 2.44 mmol/L for V. parahaemolyticus and Salmonella, respectively. BZITC inhibited tdh gene expression in a dosedependent manner. In summary, BZITC has a good effect against two gram-negative pathogens, and the presence of benzene group of isothiocyanates flavors has a certain impact on the antibacterial effect. These results suggest BZITC could be potentially applied as food preservatives.
