人参皂甙Rh_2诱导C_6胶质瘤细胞凋亡的实验研究 被引量:19

摘要 目的:观察不同浓度人参皂甙Rh2(G-Rh2)对C6胶质瘤细胞的诱导凋亡作用,并在分子水平探讨其作用机理。方法:细胞计数法检测细胞增殖,流式细胞仪分析细胞的DNA组织含量变化,电镜下观察细胞的超微结构改变。结果:1)不同浓度G-Rh22、4、8μmol/L对C6胶质瘤细胞有增殖抑制作用,且呈明显量效关系;2)流式细胞术分析经G-Rh2作用72h的C6胶质瘤细胞的DNA组织直方图提示有凋亡发生;3)不同浓度G-Rh22、4、8μmol/L可使G0/G1期细胞比率下降(P<0.05,P<0.01),S期细胞比率明显升高(P<0.05,P<0.01),而G2/M期细胞相对稳定(P>0.05);4)电镜结果提示经4μmol/LG-Rh2作用72h的C6细胞呈典型的凋亡形态学改变。结论:G-Rh2对C6胶质瘤细胞有诱导凋亡作用,呈剂量依赖性抑制C6胶质瘤细胞增殖,对脑胶质瘤的治疗有重要意义。 Objective:To investigate G-Rh 2 's function of suppressing the growth of glioma C 6 cells、inducing apoptosis and finding the mechanism of this effect on molecular level.Methods:Cell proliferation was evaluated by cell numeration,the changes of cell DNA histogram were tested by flow cytometry(FCM)and cell ultrastructures were observed by transmission electron mi croscope(TEM).Results:①G-Rh 2 exerted a dose-dependence inhibitory effect on cell prolifera tion at the dose between2~8uM.②After treated with G-Rh 2 for72h.C 6 cells were induced significant apoptosis effect in cell DNA histogram.③the proportion of G0/G1phase cell were significantly decreased,concomitant with a increase in the proportion of S phase cells.④Exposure to G-Rh 2 resulted in apoptosis changes in ultrastructure of C 6 cells.Conclusion:G-Rh 2 can in hibit proliferation and induce apoptosis of C 6 Cells.
