摘要
目的:优选四物汤补血有效部位(多糖、二氯甲烷和正丁醇部位)的最佳配比,并探讨其补血作用机制。方法:采用均匀设计法,选用U7(76)表分组设计,采用ip磷酰胺(CTX)复制小鼠骨髓造血功能抑制模型,以红细胞计数(RBC)、红细胞比容(HCT)、血红蛋白(HGB)为考察指标,筛选有效部位的最优配比。采用酶学比色法测定Na+-K+-ATP酶活性,检测血清中IL-3、EPO含量。结果 :经多元统计分析,四物汤补血有效部位最优配比为多糖部位:1 600mg/kg,二氯甲烷部位:75mg/kg,正丁醇部位:14mg/kg。配比优化方能显著提高红细胞膜Na^+-K^+-ATP酶活性,并能显著提高IL-3和EPO水平。结论:均匀设计结合药效学实验的方法可确定四物汤有效部位的最优配比。其补血作用机制与提高红细胞膜上Na^+-K^+-ATP酶活性和IL-3、EPO水平有一定相关性。
Objective:To optimize the compatibility of three effective parts(such as polysaccharide,dichloromethane and n-butanol part)for blood supplement in Siwu Decoction and explore the possible mechanism.Methods:To replicate the mouse bone marrow hematopoietic inhibition model with ip cyclophosphamide(CTX)and take RBC,HCT and HGB as evaluating indicators,then screen the best compatibility according to the table U7(76)of uniform design.To determinate the activity of Na+-K+-ATP enzyme by enzymology colorimetric method,then detect the content of IL-3 and EPO in serum.Results:According the multivariate statistical analysis,the optimal compatibility of effective parts for blood supplement in Siwu Decoction was polysaccharide part:1600 mg/kg,dichloromethane part:75 mg/kg,n-butanol part:14 mg/kg.The compatibility of effective parts can increase significantly the activity of Na^+-K^+-ATP enzyme and the content of IL-3,EPO.Conclusion:The best compatibility of effective parts can be determined by uniform design combined with pharmacodynamic experiments.The mechanism of blood enriching action may be related to raising the activity of Na^+-K^+-ATP enzyme on erythrocyte membrane and the level of IL-3,EPO.
