摘要
采用单因素与正交试验结合的方法对白芷叶总香豆素的大孔树脂纯化工艺进行研究,发现其优选工艺为:大孔树脂型号为D-101、上柱液的质量浓度为0.2 g/mL、上柱体积为3 BV,上柱液pH为4,上柱液流速为2 BV/h,洗脱用乙醇体积分数为90%,洗脱速度为2 BV/h,洗脱体积为6 BV.采用该纯化工艺,可使白芷叶总香豆素的纯度达到58.3%.
The method of combining single factor with orthogonal test was used to study the purification process of macroporous resin from total coumarin of Angelica Dahurica leaves.The optimized process is as follows:the macroporous resin model is D-101,the concentration of the sample solution is 0.2 g/mL,the volume of the supernatant is 3 BV,the pH of the sample is 4,the loading speed is 2 BV/h,the ethanol volume fraction is 90%,the elution rate is 2 BV/h and the elution volume is 6 BV.Using the optimized process,total coumarin purity of Angelica Dahurica leaves is up to 58.3%.
