摘要
目的 本研究采用人宫颈癌细胞 (He L a细胞株 ) ,观察异鼠李素诱导下 He L a细胞 bcl- 2和 bax基因表达的改变以研究其抗癌作用的机理。方法 体外培养的人宫颈癌细胞 (He L a)经 2 0μg/ml异鼠李素处理后 ,作光电镜观察 ,集落形成实验 ,3H- Td R掺入实验和流式细胞术检测。结果 异鼠李素作用后 ,He L a细胞的生长明显受到抑制 ,细胞形态出现凋亡特征 ,集落形成率和3H- Td R掺入率显著低于对照组 ,流式细胞仪检测显示异鼠李素处理组的细胞被阻止在G2 /M期 ,细胞凋亡率升高 ,bcl- 2癌基因表达降低 ,bax癌基因表达明显增加。结论 异鼠李素可能通过影响癌基因表达 ,从而抑制 He L a细胞增殖 ,达到抑制肿瘤的作用。
Aim to investigate the inhibition effect of isorhmnetin on oncogene expression of bcl 2 and study to the mechanism of inhibition effect on cancer.Methods By techniques of cell culture in vitro, HeLa cells were treated by isorhmnetin in 20 μg/ml,then observed and analyzed by light microscopy, electronic microscopy, 3H TdR incorporate assay and Flow Cytometry (FCM). Results The growth of HeLa cells was markedly inhibited. The rate of 3H TdR incorporation and colony forming capacity both declined. Flow cytometry technique demonstrated that the rate of apoptosis of those cells increased. oncogene expression of bcl 2 markedly decreased, but bax oncogene protein markedly increased. Conclusions isorhmnetin could have effect of inhibiting growth onr ocervical cancer cells.
