摘要
目的 :评价利鲁唑对吗啡躯体依赖的调节作用及可能机制。方法 :在小鼠吗啡依赖和大鼠依赖模型上分析利鲁唑对吗啡躯体依赖的影响。用化学发光法测定小鼠脑组织NOS活性。结果 :在小鼠吗啡躯体依赖模型中 ,利鲁唑 (2 .5~ 1 0mg/kg ,sc)剂量依赖性地抑制吗啡依赖小鼠纳洛酮催促的戒断症状 ;在大鼠吗啡依赖模型中 ,利鲁唑 (2 .5和 5mg/kg,sc)使吗啡依赖大鼠纳洛酮催促戒断积分明显降低。利鲁唑能显著降低吗啡依赖小鼠不同脑区(大脑、小脑、丘脑 )一氧化氮合酶 (NOS)活性。结论 :利鲁唑抑制吗啡依赖大鼠和小鼠的戒断症状 。
Objective:To study the influence and the possible mechanisms of riluzole on the physical dependence on morphine.Methods:The influence of riluzole on morphine dependence was studied in mice and rats in vivo . NOS activity was determined as a measure of concentration of NO, using a direct NOS assay kit. Results:In morphine dependent model of mice, withdrawal syndrome induced by naloxone was inhibited by co_administration of riluzole (2.5-10?mg/kg, sc) with morphine in a dose dependent manner. In morphine dependent model of rats, co_administration of riluzole (2.5 and 5?mg/kg, sc) with morphine could inhibit the rat abstinent syndrome induced by naloxone significantly. Naloxone induced an increase in NOS activity in cerebrum, thalamus and cerebellum of morphine_dependent mice, co_pretreatment of mice with morphine and riluzole inhibited this effect induced by naloxone. Conclusions:The results indicated that riluzole was able to inhibit substance dependence on morphine in mice and rats, which was related with its inhibitory action on NOS activity.
