增殖细胞核抗原与卵巢癌细胞系化学敏感性的关系

Correlation of chemosensitivity to anticancer drugs and PCNA expression in human epithelial ovarian cancer cell lines

摘要 目的 探讨增殖细胞核抗原(PCNA)与上皮性卵巢癌细胞系化学敏感性的关系。方法 使用顺为铂作用于A2780及AD60细胞株。在MTT法测定耐药倍数基础上,用ABC检测PCNA表达。结果 MTT测定AD60的耐药倍数为2.4。A2780 PCNA的表达低于AD60,且随药物浓度的递增染色较耐药组明显变浅。A2780、AD60相同药物浓度作用后PCNA的表达有显著性差异(P<0.01);同一细胞株不同药物浓度作用后PCNA表达有显著性差异(P<0.01)。结论 增殖细胞核抗原表达与上皮性卵巢癌细胞对细胞毒性药物的化学敏感性呈负相关。 Objective To evaluate the relationship between PCNA expression and chemosensitivity in epithelial ovarian cancer cell lines. Methods The human epithelial ovarian cancer drug-sensitive cell lines A2780 and drug-resistance cell lines AD60 was treated with cis-plati. MTT was used to mearure the multiple of drug resistance. PCNA expression was studied by avidin-biotin-peroxidase complex method (ABC). Results the multiple of drug resistance of AD60 was 2.4 PCNA expression become lighter with the increasing cisplatin concentration in A2780 than those in AD60. there were significant difference of PCNA expression between the two cell lines in the same cisplatin concentrations. Meanwhile, there were significant difference of PCNA expression in the same cell lines with the differencent cisplatin concentrations. Conclusion The current study suggests that PCNA expression is relate to the chemosensitivity in epithelial ovarian cancer. The higher telomerase activity and PCNA expression the lower chemosensitivity in cancer cells.
