摘要
研究了茶树芽叶蛋白质提取纯化及双向电泳技术,在样品制备、电泳及染色等方面进行了技术改进,解决了茶树芽叶中富含色素、多酚类化合物及其氧化产物等对双向电泳严重干扰的问题,探索出一种可获得重复性好,清晰度高的蛋白质双向电泳图谱技术;同时发现一种辨别茶树蛋白质样品质量好坏的简便方法。电泳图谱可分辨出茶树芽叶蛋白质斑点数500个左右,蛋白质相对分子量约在14.0 kD~100.0 kD范围内,主要分布在14.0 kD~67.0 kD之间;等电点约在4~9.5范围内,主要分布在4~6.5。
The technology of two-dimensional electrophoresis(2-DE) of protein from tea plant including the preparation of protein sample, the electrophoresis and staining was improved. The interference of rich tea pigments, polyphenols and their oxides on the stability and clearness of two-dimensional electrophoresis was solved. At the same time, a simple method which can verify the quality of protein sample from tea plant has been found. About 500 protein spots could be visualized on the 2-DE map by staining. Their relative molecular weight of protein was between 14.4 kD~100.0 kD,most of them were in the range of 14.0 kD~67.0 kD. The isoelectric points were found to lie between 4.0~9.5,most of them were in the range of 4.0~6.5.
