β-巯基乙醇诱导成年鼠骨髓基质细胞向神经细胞分化的研究 被引量:4

摘要 目的 :探讨β 巯基乙醇诱导成年大鼠骨髓基质细胞成神经细胞的可能性。为神经再生和诱导剂的筛选提供一种新的参考方法。方法 :从大鼠骨髓中分离出骨髓基质细胞并培养传至 3- 5代 ,诱导前 2 4h用含 1mmol /l的β 巯基乙醇及含 2 0 %的DMEM培养液培养 ,再以含 5mmol /lβ 巯基乙醇的无血清DMEM培养液作诱导 ,用免疫细胞化学方法鉴定神经细胞特异性标志物NF、NSE及GFAP的表达。结果 :诱导后 1h ,部分细胞发生形态学上的改变 ,6h后大部分细胞不仅在形态上表现为神经元样特征 ,而且NF、NSE等特异性抗体呈阳性表达 ,而GFAP抗体表达呈阴性。结论 :β Objective:To study the possibility thatβ mercaptoethanol induces adult rat bone marrow stromal cells(BMSCs)into neurons for further investigation of an alternative for neurons regeneration and the screening of inducers.Methods:The bone marrow stromal cells were isolated from adult rats and cultured for 3~5 passages.The BMSCs were incubated with DMEM containing 20% FBS and 1 mmol/l β mercaptoethanol (β ME) for 24 hours.Then the media were replaced by DMEM media consisting of 5 mmol/l β mercaptoethanol.The morphological changes of the cells were observed under phase contrast microscope,the cells were stained immunocytochemically with NF,NSE,and GFAP antibodies,respectively.Results:After an hour of induction with β ME,some cells showed morphological changes.Six hours later,many BMSCs became neuron like cells and were positively stained with NSE and NF antibodies,but negatively with GFAP.Conclusion:The bone marrow stromal cells can differentiate into neuron like cells as induction by β mercaptoethanol.
