摘要
采用小鼠活动实验及小鼠自由活动脑电描记实验。小鼠腹腔注射马钱子碱350μg/kg,产生兴奋、活动量增加。而14~23mg/kg小鼠出现明显镇静、闭目、活动量减少。小鼠腹腔注射23mg/kg后,监测描记脑电、肌电6h,总效应是缩短入睡时间、减少觉醒时间及延长总睡眠时间,与自身对照有较显著差异(P<0.05)。结果提示:马钱子碱对小鼠兼有兴奋与抑制两种作用。两种作用的出现常取决于药物的剂量及动物个体对药物的敏感性。
The purpose of the experiment is to prove the effects of brucine on the central nervous system of mice. ① The intraperitoneal injection of 350μg/kg of brucine to mice can produce reflex responses whereby the motor component of the reflex is augmented and the inhibitory component diminished. The effects mentioned above are similar to those of 300μg/kg strychnine IP. 14~23mg/kg brucine IP to mice can produce sedation, closing eyes and diminishing body movement, ② The electroencephalographic activities in mice were studied in the experimental'sleeping box' in which the mice were given 23mg/kg of brucine, and kept for 6 hours. During the period its sleep patterns were monitored by electroencephalographic and electromyographic recordings. These experiments showed that their overall effects were to shorten the duration of falling asleep and increase the total sleeping time. A characteristic changes in sleep pattern appeared to be an augmentation of the duration of slow wave as compared with the self control (P<0.01, n=4). This study showed that brucine had both depressing and stimulating actions and the specific effects observed depended on the dose used.
