摘要
目的:通过网络药理学的方法初步探讨射干麻黄汤治疗支气管哮喘的作用机制。方法:基于TCMSP、Pubchem、BATMAN-TCM数据库获取射干麻黄汤化学成分及其对应靶点,基于OMIM、Drugbank、Disgenet数据库获取支气管哮喘相关靶点,将药物靶点与疾病靶点映射得到的重复靶点视为射干麻黄汤作用于支气管哮喘的预测靶点,利用Cytoscape软件构建中药-靶点及蛋白相互作用网络(PPI),对PPI网络进行拓扑分析得到Hub蛋白,通过Reactome对预测靶点进行通路富集分析。结果:共获得95个化合物,219个射干麻黄汤验证靶点及513个预测靶点和224个支气管哮喘相关靶点,映射后获得54个射干麻黄汤作用于支气管哮喘的预测靶点,拓扑分析得到NR3C1、HPS90AA1、PARP1、TGFB1等9个Hub蛋白。Reactome信号通路共239条,其中14条显著富集,主要涉及免疫、信号转导、代谢3种生物途径。HPS90AA1、TGFB1、NFKB2、MMP9可能是射干麻黄汤作用于免疫性炎症通路的关键靶点,NR3C1、PGR可能是其治疗难治性哮喘的关键靶点。结论:本研究系统地探讨了射干麻黄汤治疗支气管哮喘的作用机制,为今后进一步实验验证提供了依据。
Objective:To investigate the mechanism of Shegan Mahuang Decoction for treatment of bronchial asthma based on network pharmacology.Methods:Based on the TCMSP database,Pubchem and BATMAN-TCM database,the chemical compositions of Shegan Mahuang Decoction and corresponding targets were obtained.The targets of bronchial asthma were obtained from the OMIM,Drugbank and Disgenet database.Then the targets of Shegan Mahuang Decoction were mapped to the targets of bronchial asthma.The herb-target network and protein-protein interaction(PPI)network were constructed using the Cytoscape software.The hub proteins of the PPI network were obtained by network topological analysis.Reactome was used to explore the potential pathways affected by Shegan Mahuang Decoction.Results:Totally 95 components,219 validated targets and 513 predicted targets of Shegan Mahuang Decoction as well as 224 targets of bronchial asthma were obtained.A total of 54 targets were obtained from the overlaps of the targets of Shegan Mahuang Decoction and bronchial asthma.The key targets were NR3 C1,HPS90 AA1,PARP1,TGFB1,etc.There were 239 Reactome pathways and 14 of them were obviously enriched.And these pathways involved 3 biological pathway,including immune,signal transduction and metabolism.HPS90 AA1,TGFB,NFKB2 and MMP9 may be the key targets for Shengan Mahuang Decoction acting on the immune inflammatory pathways.NR3 C1 and PGR may be the key targets for treatment of refractory asthma.Conclusion:This study systematically discussed the mechanism of Shegan Mahuang Decoction for treatment of bronchial asthma.It provides a theoretical basis for further experimental verification of the mechanism of Shegan Mahuang Decoction.
