Expression of HSP90α in hepatocellular carcinoma and its significance

摘要 目的 研究热休克蛋白 90α(HSP90α)在人原发性肝细胞癌 (HCC)组织中的表达并探讨其与肿瘤生物学行为、预后的关系。方法 采用免疫组织化学方法检测了 10例正常肝组织、40例HCC肿瘤组织及癌旁组织中HSP90α的表达。结果 HSP90α在正常肝脏、癌旁组织、原发性肝癌中的阳性表达率分别为 10 %、5 2 5 %、72 5 %。原发性肝癌中的阳性表达率明显高于正常肝脏和癌旁组织 (P <0 0 5 )。HCC的HSP90α与肿瘤分化程度、肿瘤大小显著相关 (P <0 0 5 ) ;与病灶数目无关(P >0 .0 5 ) ;与预后亦有关 ,HSP90α阴性病人预后显著优于HSP90α阳性病人 ,术后平均无瘤生存期前者 38 6个月、后者 2 5 5个月 (P <0 0 5 )。结论 HSP90α在原发性肝癌中表达增高 ,HSP90α可作为判断HCC分化程度和预后的指标。 Objective To investigate the expression of HSP90α in human hepatocellular carcinoma (HCC) and explore its relation to biological behavior and prognosis of the tumor. Methods Immunohistochemical S-P method was used to detect the expression of HSP90α in liver tissue from 10 normal individuals and liver and juxtacancerous tissues from 40 patients with HCC. Results The expression rate of HSP90α was 10%, 52.5% and 72.5% in normal liver tissue, juxtacanerous tissue and HCC tissue, respectively. A significantly higher distribution of HSP90α expression in HCC tissue compared with juxtacancerous tissue and normal liver tissue was found (P<0.05). HSP90α was markedly correlated to tumor differentiation and size (P<0.05) but not to tumor number (P>0.05). Meanwhile, it was correlated with prognosis of HCC. The mean tumor-free survival time in HSP90α-negative patients was 38.6 months while that in HSP90α-positive ones 25.5 months. There was significant difference between these 2 groups of patients (P<0.05). Conclusions HSP90α is overexpressed in human HCC and can be used as an indicator to judge differentiation and prognosis of HCC.
