摘要
目的研究硫酸铵梯度法制备盐酸环丙沙星脂质体的影响因素。方法采用阳离子交换树脂-一阶导数分光光度法测定包封率 ,考察硫酸铵的作用及温度、药脂比对包封率的影响。结果在各种梯度法制备的盐酸环丙沙星脂质体中 ,硫酸铵梯度法得到的包封率可达 92 4 % ;盐酸环丙沙星脂质体的包封率随着硫酸铵浓度和温度的升高而增加 ,随着药脂比的增大而降低。结论硫酸铵梯度是盐酸环丙沙星脂质体包封率得以提高的主要原因 ,硫酸铵浓度。
Objective To study the influencing factors on preparation of ciprofloxacin into liposomes by ammonium sulfate transmembrane gradients.Methods The entrappent efficiencies of ciprofloxacin liposomes were determined by a cation exchange resin-first order derivative spectrophotometry method,and the influence of various factors on the entrappent efficiencies was appreciated.Results The entrappent efficiency of ciprofloxacin liposomes prepared by the ammonium sulfate gradients method is 92.4%,much higher than that with the other gradients.The entrappent efficiency was enhanced by the increased temperature,the increased ammonium sulfate concentration and the decreased drug-to-lipid ratio.Conclusions The preparation of ciprofloxacin into liposomes is dependent on the gradients of ammonium sulfate,both the ammonium cation and the sulfate anion play important roles in the drug loading.In the process of loading,the temperature and drug-to-lipid ratio are important to the entrappent efficiency.
