摘要
目的:体外无血清条件下诱导人脐带间充质干细胞向肝样细胞分化。方法:在无血清条件培养体系下,利用肝细胞生长因子和致瘤素联合诱导人脐带间充质干细胞向肝样细胞分化。观察细胞形态学变化,检测诱导细胞甲胎蛋白、细胞角蛋白-18、白蛋白mRNA及相应蛋白的表达及糖原储存情况。结果:经肝细胞生长因子、致瘤素联合诱导后,细胞形态由长梭形变为类圆形,表达甲胎蛋白、细胞角蛋白-18、白蛋白mRNA及相应蛋白,具有合成和储存糖元的功能。结论:体外无血清培养扩增人脐带间充质干细胞,经肝细胞生长因子(20ng/ml)与致瘤素(20ng/ml)联合诱导后,初步具有向肝样细胞分化的能力。
Objective:To induce human umbilical cord mesenchymal stem cells into hepatocyte-like cells under serum-free condition in vitro.Methods:The cells treated with serum-free medium,containing 20ng/ml hepatocyte growth factor(HGF)and 20ng/ml oncostatin(OSM).Cells was observed with phase contrast microscope.Expressions of alpha fetoprotein,albumin and cytokeratin18 were analyzed by immunocytochemistry,similar to the findings of the expressions of AFP,ALB and CK18 mRNA.The glycogen deposits were examined by periodic acid-Schiff method.Result:The morphology of hUCMSC is the same as typical fibroblast.hUCMSC kept a capability of multiplication.hUCMSC could be induced toward osteoblast and adipocyte under 3 weeks’ appropriate experimental conditions.MSCs were induced into hepatocyte-1ike cells that is positive for AFP,ALB and CK18.Cellolar morphology changed from a spindle to a rather polygonal shape.Induced cells were positive for glycogen staining.Conclusion:The human umbilical cord mesenchymal stem cells could be differentiated into heaptocyte-like cells in serum-free medium with the existence of 20ng/ml HGF and 20ng/ml oncostatin.
