血小板与血管内皮下层粘附的体外实验模型 被引量:1

摘要 报道了一种研究血小板与血管壁内皮下层粘附的体外实验模型。血管壁经1.0%NP-40处理5min,300mmo/L氯化钾处理10min,内皮剥脱。血小板用吖啶橙标记,血小板内可见橙黄色颗粒。血管壁与血小板反应后,扫描电镜观察可见内皮下层表面有大量血小板粘附。用2.0%SDS洗脱粘附的血小板,用荧光分光光度计测定洗脱血小板的荧光强度,结果表明,吖啶橙标记后血小板浓度与其荧光强度成正比。粘附血小板个数用下式计算: An experimental model in vitro of the adhesion of platelets with vessel sube-ndothelium is reported. The vessel wall was treated with 1.0% Nonidet P-40 for 5min and with 300mmol/L KC1 for 10 min. The whole endothelium was removed. The platelets were labeled with acridine orange. Orange granules were observed in labeled platelets under fluorescence microscope. Under scanning electron miceo-scope, a great number of adhered platelets were seen on the surface of the suben-dothelium following the vessel wall reacting with platelets. The adhered platelets were washed away with 2.0% SDS solution. The intensity of fluorescence of the washed platelets was measured with a spectrofluorophotometer. The result indicates that the concentration of platelets labeled with fluorescein is in direct proportion to their intensity of fluorescence. The number of the adherent platelets on each specimen was calculated according to the following formula: Number of adherent platelets =platelet concentration of standard tube/ intensity of florescence of standard tube k sample volume in the test tube × intensity of fluorescence of the sample.
