人参皂苷Rg_1及其肠内菌代谢产物Rh_1对小鼠免疫细胞功能的影响 被引量:50

摘要 目的 初探人参皂苷Rg1 及其肠内菌代谢产物Rh1 对正常小鼠免疫功能的影响。方法 用Rh1 与Rg1 分别处理脾T细胞 ,B细胞及腹腔巨噬细胞 (Mφ) ;MTT比色法测T和B细胞增殖能力 ;中性红比色法测Mφ的吞噬功能 ;Griess法测Mφ释放NO的水平。 结果 Rh1 能促进脾细胞增殖、下调ConA诱导的T细胞增殖 ;Rh1 与Rg1 对LPS诱导的B细胞增殖均无明显作用 ;Rg1 和Rh1 能提高Mφ的吞噬能力和促进NO的释放。 结论 Rg1 及其代谢产物Rh1 可共同作用于T细胞和Mφ而产生免疫调节作用。 AIM To study the mechanism of action of ginsenoside Rg 1 and its metabolite Rh 1 on the function of immunocyte in normal mice. METHODS The proliferation of splenocytes was determined by using MTT assay. Phagocytosis activity of peritoneal macrophages was determined by using neutral red phagocytosis assay. Griess agent was used to assay the NO-releasing level of peritoneal macrophages. RESULTS Rh 1 was shown to promote proliferation of splenocytes directly, down-regulate the Con A-induced proliferation of T-lymphocytes. Rg 1 can not induce proliferation of splenocytes directly, while Rg 1 100 μg·mL -1 can inhibit ConA-induced proliferation of T-lymphocytes obviously. Both Rh 1 and Rg 1 showed no effect on LPS-induced proliferation of B-lymphocytes. Rg 1 and Rh 1 were found to significantly increase the phagocytosis and NO-releasing level of peritoneal macrophages, while the effect of Rh 1 is weaker than that of Rg 1. CONCLUSION The immuno-regulation effect of Rg 1 may be carried out by both Rg 1 and its metabolite Rh 1. Their functions on other immunocytes still need further study.
