日粮添加染料木黄酮对产蛋后期种鸡子代骨骼肌发育的影响

Effects of Dietary Genistein Supplementation for Broiler Breeder Hens on Skeletal Muscle Development of Offspring

摘要 试验旨在研究日粮添加染料木黄酮(genistein,GEN)对种鸡子代骨骼肌发育的影响。选用产蛋后期种母鸡180只随机分为3组,对照组饲喂玉米-豆粕型基础日粮,试验组分别在基础日粮中添加GEN 20 mg/kg和40 mg/kg。种鸡饲喂第28天后,每处理收集80枚种蛋孵化。对应种鸡分组将子代肉仔鸡分为3个处理组,即CON组,L-GEN组和H-GEN组,饲喂相同基础日粮至28日龄。结果显示:与CON组相比,L-GEN组和H-GEN组均显著提高13胚龄鸡胚重、体长、相对胚重和相对体长(P<0.05);显著提高子代1、8、15日龄和22日龄仔鸡体重,以及1日龄和28日龄仔鸡胸肌重(P<0.05)。组织学分析结果表明,GEN处理显著提高子代肉仔鸡1日龄胸肌纤维密度和28日龄胸肌纤维直径(P<0.05)。qRT-PCR分析表明,13胚龄时,L-GEN组和H-GEN组胸肌中MSTN的mRNA表达水平较CON组显著降低(P<0.05),而IGF-1R和Myf5的表达水平显著提高(P<0.05);L-GEN组胸肌ERα的mRNA表达水平显著提高(P<0.05),H-GEN组胸肌MyoG的表达水平显著提高(P<0.05)。1日龄时,L-GEN组和H-GEN组仔鸡胸肌中ERα、IGF-1R、MyoD和mTOR的mRNA表达水平较CON组显著提高(P<0.05),MuRF1和Atrogin-1的表达水平则显著降低(P<0.05);L-GEN处理显著提高胸肌中MyoG的mRNA表达水平(P<0.05);H-GEN组胸肌中ERβ、Myf5和Pax-7的mRNA表达水平显著提高(P<0.05)。结果提示,产蛋后期种鸡日粮中添加GEN能够促进子代骨骼肌发育,提高生长性能。 This study aimed to investigate the effects of dietary genistein(GEN)supplementation in later-phase breeder hens on skeletal muscle development of offspring.A total of 180 healthy 60-week-old Qiling breeder hens with similar body weight and egg production rate were ran⁃domly divided into 3 groups.The control group(CON)was fed a corn-soybean meal type basal diet,while the treatment groups were supplemented with 20 and 40 mg/kg GEN in the basal diet,respec⁃tively.On day 28 of the breeder diet treatment,80 fertilized breeding eggs were randomly collected from each treatment and incubated using conventional procedures.The offspring were divided into three treat⁃ment groups according to the breeder′s diet:CON,L-GEN and H-GEN groups.At 13 embryonic days,a sample of chicken embryos was collected once for indicator testing.After fledge,1-day-old male chicks were selected and fed the same diet until 28 days of age.The results showed that both the L-GEN and HGEN groups significantly increased the embryonic weight,body length,relative embryonic weight and rela⁃tive body length of the 13-embryo old chicks(P<0.05);significantly increased the body weights of the off⁃spring at 1,8,15 and 22 days of age(P<0.05);significantly increased pectoral muscle weights of the off⁃spring at 1 and 28 days of age(P<0.05).Histological analysis showed that maternal supplementation with GEN significantly increased pectoral muscle fiber density at 1 day of age and pectoral muscle fiber diameter at 28 days of age(P<0.05).qRT-PCR results showed that at 13 embryonic ages,compared with CON group,the mRNA expression of IGF-1R and Myf5 significantly up-regulated and the mRNA expression level of MSTN down-regulated in skeletal muscle of offspring chicks in the L-GEN and H-GEN groups(P<0.05);L-GEN treatment significantly increased the mRNA expression of ERαin the skeletal muscle(P<0.05);HGEN treatment significantly increased the mRNA expression of MyoG in the skeletal muscle(P<0.05).At 1 day of age,the mRNA expression of ERα,IGF-1R,MyoD and mTOR in the skeletal muscle of broiler off⁃spring was significantly up-regulated in the L-GEN group and the H-GEN group(P<0.05)and the mRNA expression of MuRF1 and Atrogin-1 were down-regulated(P<0.05);the mRNA expression of MyoG in the skeletal muscle was significantly increased in the L-GEN group(P<0.05);the mRNA expression of ERβ,Pax-7 and Myf5 in the skeletal muscle was significantly increased in the H-GEN group(P<0.05).The results indicated that dietary genistein supplementation could promote skeletal muscle development in the offspring of broilers,thereby improving the growth performance.
