火针对免疫抑制模型大鼠的血常规和IL-2、IL-6的影响

Effect of Fire Needling on Blood Parameters,IL-2 and IL-6 in Immunosuppressed Rat Models

摘要 目的 旨在探究火针治疗对环磷酰胺(Cyclophosphamide, CTX)所致免疫抑制模型大鼠的免疫功能的影响,并分析其潜在的作用途径。方法 将SPF级健康雌性Wistar大鼠32只,采用随机数字表法分为空白组、模型组、针刺组、火针组,每组8只。其中模型组、针刺组、火针组分别建立免疫抑制大鼠模型。针刺组、火针组造模后分别对中脘、足三里两穴进行治疗。治疗后对大鼠外周血样本进行血常规检测,并对白细胞介素2(Interleukin-2,IL-2)、白细胞介素6(Interleukin-6,IL-6)的水平进行定量分析。结果 与模型组比较,干预后,火针组白细胞计数水平上升,针刺组、火针组干预后白细胞计数水平均高于干预前,结果均有统计学差异(P<0.05);与空白组比较,模型组IL-2含量降低,具有差异性(P<0.05)。与模型组相比,针刺组、火针组的IL-2平均水平明显高于模型组,具有统计学差异(P<0.05)。与模型组比较,针刺组的IL-6平均水平降低,其结果具有显著性差异(P<0.01)。结论 火针可以提高CTX免疫抑制大鼠模型的白细胞计数水平,并且可以调节IL-2、IL-6细胞因子水平,提高辅助性T淋巴细胞1(T helper cell 1,Th1)细胞功能,促进Th1与辅助性T淋巴细胞2(T helper cell 2,Th2)向平衡状态发展,增强机体的细胞免疫功能。 Objective The present study aimed to investigate the effects of fire needling on the immune function of immunosuppressed rats induced by cyclophosphamide(CTX)and analyze its potential mechanisms of action.Methods Thirty-two healthy female Wistar rats were randomly divided into four groups(with eight rats/in each group):a blank group,a model group,a needling group,and a fire needling group.The immunosuppressed rat models were established in the model group,the needling group,and the fire needling group.After modelling,the needling group and fire needling group received treatment at the Zhongwan(CV12)and Zusanli(ST36)acupoints.Subsequently,peripheral blood samples of the rats were analyzed for hematological parameters,and the levels of interleukin-2(IL-2)and interleukin-6(IL-6)were quantitatively determined.Results Compared with the model group,the white blood cell(WBC)count in the fire needling group increased after intervention,and the WBC count in the needling group and fire needling group was higher than that before intervention,with statistically significant differences(P<0.05).Compared with the blank group,the IL-2 content in the model group was significantly lower(P<0.05).Compared with the model group,the average level of IL-2 in the needling group and fire needling group was significantly higher(P<0.05),and the average level of IL-6 in the needling group was lower,with a statistically significant difference(P<0.01).Conclusion Fire needling can improve the WBC count in the CTX-induced immunosuppressed rat model and regulate the levels of IL-2 and IL-6 cytokines.It can enhance T helper cell 1(Th1)function,promote the balance between Th1 and T helper cell 2(Th2),and enhance the cellular immune function of the body.
