左右半结肠癌患者肠道菌群构成差异分析

摘要 目的:探讨左、右半结肠癌患者肠道菌群组成差异。方法:收集诊断为结直肠癌并完成手术的28例癌组织及癌旁组织资料,其中左半结肠癌18例,右半结肠癌10例,并对2组患者年龄、性别、身高、体质量、身体质量指数(BMI)及体型进行比较。利用16S rRNA测序技术测定每个样本的菌群种类及丰度,分别比较左半结肠癌与右半结肠癌组织、左半结肠癌与其癌旁组织、右半结肠癌与其癌旁组织、左右半结肠癌癌旁组织中菌群分布的差异。结果:左右半结肠癌患者年龄、性别、身高、体质量、BMI及体型比较差异无统计学意义(P>0.05);在门水平上,左右半结肠癌组织菌群进行比较,右半结肠癌组织中丰度较高的菌群是互养菌门,左半结肠癌组织中丰度较高的菌群是疣微菌门、绿弯菌门、软壁菌门,差异有统计学意义(P<0.05);在属水平上,左右半结肠癌组织菌群进行比较,左半结肠癌组织中菌群丰度明显高于右半癌组织(P<0.05),其中右半结肠癌组织中丰度较高的菌群有月形单胞菌属、小单胞菌属、布雷德菌属、消化球菌属,左半癌组织丰度较高的菌群有阿克曼菌属、沙雷氏菌属、瘤胃球菌属、芽胞杆菌属、双歧杆菌属、韦荣球菌属、毛螺旋菌属、鞘氨醇单胞菌属、克雷伯氏菌属、厌氧杆菌属、寡养单胞菌属,差异有统计学意义(P<0.05)。结论:左右半结肠癌组织中菌群丰度存在明显差异,可能是左右半结肠癌的发生、发展及预后差异的原因之一。 Objective:To investigate the difference in intestinal flora composition between patients with left and right-sided colon cancer.Methods:Colonic tumor tissues and adjacent tissues were collected from 28 patients diagnosed with colorectal malignancy who underwent surgery,including 18 cases of left colon cancer and 10 cases of right colon cancer.Age,sex,height,weight,BMI,and body type of the two groups were compared.16S rRNA sequencing technology was used to determine the flora species and community richness of each sample.The differences in flora distribution were compared between the left colon cancer and right colon cancer tissues,left colon cancer and its adjacent tissues,right colon cancer and its adjacent tissues,and the left and right colon cancer and its adjacent tissues.Results:There were no statistically significant differences between patients with left and right-sided colon cancer in terms of age,sex,height,weight,BMI,and body type(P>0.05).At the Phylum level,the microbial comparison between the left and right-sided colon cancer tissues revealed that Synergistetes had a higher community richness in the right-sided cancer tissues.Verrucomicrobia,Chloroflexi,and Tenericutes had a higher community richness in the left-sided cancer tissues with statistically significant differences(P<0.05).At the Genus level,the microbial community richness in the left-sided cancer tissues was significantly higher than in the right-sided cancer tissues(P<0.05).Selenomonas,Parvimonas,Bulleidia,and Peptococcus had a higher community richness in the right-sided cancer tissues,and Akkermansia,Serratia,Ruminococcus,Bacillus,Bifidobacterium,Veillonella,Lachnospira,Sphingomonas,Klebsiella,Anaerostipes,and Stenotrophomonas had a higher community richness in the left-sided cancer tissues with statistically significant difference(P<0.05).Conclusion:There are significant differences in the microbial community richness between the left and right-sided colon cancer tissues.The distinction in the microbial populations may also contribute to the differences in pathogenesis,progression,and prognosis between the left and right-sided colon cancer.
