血管内皮损伤相关circRNAs与复发性流产的关联性及其调控机制研究

摘要 目的通过建立反复自然流产(RSA)小鼠模型,研究血管内皮损伤相关circRNAs与复发性流产的关联性及其调控机制。方法选取8只正常妊娠模型的孕鼠设为对照组,将8只复发性流产模型的孕鼠设为实验组。基于VEGF及生物信息学构建ceRNA网络,对miRNA-VEGF和circRNAs调控网格在RSA中的作用和机制进行分析,对比两组小鼠circRNA-miRNA的相对量,分析目标circRNA-miRNA及影响因素的非条件Logistic回归结果。结果目标circRNA-miRNA在两组之间的相对量表达,差异具有统计学意义(P<0.05),miR-10a-5p在实验组中低表达,而circ-0014243、circ-0126991在实验组中高表达。通过对血管内皮损伤相关circRNA与miRNA进行研究分析,经过相对表达量的分析发现,miR-497-5p、miR-10a-5p、circ-0014243、circ-0126991比较,差异有统计学意义(P<0.05),而circ-0041000比较,差异无统计学意义(P>0.05)。结论基于构建ceRNA调控网络、RT-PCR及细胞及动物层面转染circRNA及miRNA进行验证,从而明确VEGF在RSA中的潜在机制,以此为临床治疗提供潜在靶标。 Objective To establish a mouse model of recurrent spontaneous abortion(RSA)and to study the correlation of circRNAs associated with vascular endothelial injury and recurrent abortion and its regulatory mechanism.Methods 8 pregnant mice with normal pregnancy model were selected as the control group,and 8 pregnant mice with recurrent abortion model were selected as the experimental group.The ceRNA network was constructed based on VEGF and bioinformatics,and the roles and mechanisms of miRNA-VEGF and circRNAs regulatory grid in RSA were analyzed,and the relative amounts of circRNA-miRNA in the two groups of mice were compared.Non-conditional Logistic regression results of target circRNA-miRNA and influencing factors were analyzed.Results The relative expression of the target circRNA-miRNA between the two groups was statistically significant differences(P<0.05).The expression of miR-10a-5p was low in the experimental group,while circ-0014243 and circ-0126991 were high in the experimental group.Through the study and analysis of circRNA and miRNA associated to vascular endothelial injury,the relative expression levels of miR-497-5p,miR-10a-5p,circ-0014243,circ-0126991 were compared,and the differences were statistically significant(P<0.05),while there was no significant difference in circ-0041000(P>0.05).Conclusion Based on the construction of ceRNA regulatory network,RT-PCR,and transfection of circRNA and miRNA at the cell and animal levels,the potential mechanism of VEGF in RSA was identified,so as to provide potential targets for clinical treatment.
