摘要
小檗碱(berberine, BBR)作为药用价值显著的中药活性成分,临床应用受限于溶解度低、生物利用度差等问题。为了克服这些难题,常采用化学改性或纳米载体递送的方法进行改善。然而,纳米载体递送虽有优势,却也伴随着药物过早泄漏、载药量受限及潜在毒性等不容忽视的问题。无载体纳米药物作为一种绿色、安全、简便的递送策略受到广泛关注,这种方法不仅避免了传统纳米载体的潜在风险,更以出色的高载药能力和优异的生物相容性弥补了纳米载体递送的不足。BBR作为具备强大自组装能力和卓越药理作用的中药活性成分,其无载体自组装纳米药物的研发备受瞩目。本文旨在总结BBR无载体自组装纳米药物的组合策略及其在实际应用中的表现,期望进一步推动中药现代化和新药开发的进程。
Berberine(BBR),as an active ingredient of traditional Chinese medicine with significant medicinal value,is limited in clinical application due to low solubility and poor bioavailability.Chemical modification or nanocarrier delivery methods are often used to overcome these challenges.However,despite the advantages of nanocarrier delivery,it is also accompanied by problems such as premature drug leakage,limited drug loading,and potential toxicity.Carrier-free nanomedicine has attracted extensive attention as a green,safe and simple delivery strategy,which not only avoids the potential risks of traditional nanocarriers,but also makes up for the shortcomings of nanocarrier delivery with excellent high drug loading capacity and excellent biocompatibility.The research and development of carrier-free self-assembly nanomedicine products of BBR,as an active ingredient of traditional Chinese medicine with strong self-assembly ability and excellent pharmacological effects,has attracted much attention.The purpose of this article is to summarize the combination strategy of BBR carrier-free self-assembled nanomedicine and its performance in practical application,hoping to further promote the modernization of traditional Chinese medicine and the development of new drugs.
